摘 要:为建立秋水仙素诱导桑树组织形成四倍体桑树的诱导体系,选用传统桑树品种沙2×伦109,以桑树愈伤组织、萌发种子和绿枝作为诱导材料,筛选秋水仙素诱导桑树愈伤组织形成四倍体桑树的适宜质量浓度,以及秋水仙素诱导桑树萌发种子和绿枝形成四倍体桑树的最佳质量浓度和浸泡处理时间。结果表明,秋水仙素质量浓度为10 mg/L时诱导桑树愈伤组织形成四倍体桑树的效果最好,诱导率达到2.67%;秋水仙素质量浓度为5 mg/L、浸泡萌发种子48 h诱导形成四倍体桑树效果最好,诱导率达到29.00%;秋水仙素质量浓度为15 mg/L、浸泡绿枝24 h诱导形成四倍体桑树效果最好,诱导率达到16.67%。适宜的秋水仙素质量浓度和浸泡处理时间对培育四倍体桑树品种及桑树多倍体育种具有重要意义。In order to establish the induction system of colchicine‑induced mulberry tissue to form tetraploid mulberry,the traditional mulberry variety Sha 2×Lun 109 was selected,and the callus,germinated seeds and green branches of mulberry were used as induction materials to screen the suitable concentration for colchicine‑induced mulberry callus to form tetraploid mulberry,as well as the optimal concentration and soaking treatment time for colchicine‑induced mulberry germinated seeds and green branches to form tetraploid mulberry.The results showed that the effect of inducing mulberry callus to form tetraploid mulberry was the best when the concentration of colchicine was 10 mg/L,and the induction rate reached 2.67%.When the concentration of colchicine was 5 mg/L and the germination seeds were soaked for 48 h,the effect of inducing tetraploid mulberry was the best,and the induction rate reached 29.00%.When the concentration of colchicine was 15 mg/L and the green branch was soaked for 24 h,the effect of inducing tetraploid mulberry was the best,and the induction rate reached 16.67%.Appropriate colchicine concentration and soaking time are of great significance for the cultivation of tetraploid mulberry varieties and polyploid breeding.
