摘 要:目的通过动物实验比较在血管靶向光动力疗法(vascular-targeted photodynamic therapy,V-PDT)中采用波长510 nm或532 nm激光作为激发光源的靶血管封闭效应。方法(1)比较V-PDT血管封闭效应的作用程度和作用深度。根据激发光源波长分为510 nm组和532 nm组,每组12只莱亨鸡,共24个光斑。按每千克体重5 mg药量经静脉注射光敏剂血卟啉注射液(hematoporphyrin derivative,HpD)后即刻行激光照射,功率密度100 mW/cm2,能量密度120 J/cm^(2),照光时间1200 s。于V-PDT前和V-PDT后的每日肉眼观察鸡冠颜色和表面变化,并于V-PDT后3 d行组织病理学检查,评价V-PDT血管封闭效应的作用程度和作用深度。(2)比较V-PDT血管封闭效应的持续时间。分组与V-PDT处理同上,采用光学相干断层扫描血管成像系统(optical coherence tomography angiography,OCTA)在体检测鸡冠血管密度,根据照射后鸡冠复色和血管密度恢复升高出现的时间,评价血管封闭效应持续时间。(3)给药后不同时间行V-PDT照光,比较两种波长激光作为激发光源可产生血管封闭效应的有效作用时长。鸡冠两侧面分别设四个光斑,在HpD给药后不同时间(0 h、2 h、4 h和6 h)开始行波长510 nm或532 nm激光照射,按(1)中的方法观察血管封闭效应。V-PDT后出现鸡冠复色和血管密度恢复升高的给药-照光时间间隔越长,其有效作用时长越长。结果510 nm组V-PDT效应分级显著高于532 nm组(P<0.05)。510 nm组均表现为中至重度反应(占100.0%),而532 nm组仅66.7%表现为中至重度反应,其余33.3%为轻度反应。510 nm组可观察到血管封闭效应的深度显著大于532 nm组(P<0.05),其血管封闭效应持续时间也更长。从产生鸡冠褪色效应和血管封闭效应的给药-照光时间间隔上来看,波长510 nm在更长的给药-照光时间间隔时仍能产生有效V-PDT效应。结论在相同HpD给药量和光剂量下,以波长510 nm激光为激发光源组的V-PDT血管封闭效�Objective To compare the vascular closure effect of 510 nm or 532 nm laser as light sources in vascular-targeted photodynamic therapy(V-PDT).Methods(1)Comparison of the intensity and the depth of the V-PDT action.In the 510 nm group and 532 nm group(12 Leghorn cocks with 24 spots in each group),hematoporphyrin derivative(HpD)as a photosensitizer was injected intravenously with a dosage of 5 mg/kg,and laser irradiation was given immediately after the administration with a power density of 100 mW/cm^(2) and an energy density of 120 J/cm2 for 1200 s.The color and surface changes of the combs were observed macroscopically and light-microscopically before the V-PDT and 3 days after the V-PDT to evaluate the intensity and the depth of the V-PDT action.(2)Comparison of the lasting time of vascular closure induced by V-PDT.Grouping and V-PDT processing is the same as the method in(1).Comb blood vessel density was detected with optical coherence tomography angiography(OCTA)in vivo,and the lasting time of vascular closure effect was evaluated according to the time of comb recolor and blood vessel density recovery after the Y-PDT.(3)Comparison of the effective duration of action by evaluating the vascular closure effect of V-PDT given at different time intervals after the photosensitizer administration.One Leghorn cock was used with four light spots set on each side of the comb.To compare the effective acting duration of 510 nm laser and 532 nm laser,the vascular closure effect of V-PDT was observed at four drug-irradiation time intervals(0 h,2 h,4h and 6 h)respectively according to the method in(1).The longer the drug-irradiation time interval at which the recolor and blood vessel density of comb recovered,the longer the effective duration of action.Results The photodynamic intensity was significantly higher in the 510 nm group than in the 532 nm group(P<0.05).The percentage of moderate to severe reactions was 100.0%in the 510 nm group and 66.7%in the 532 nm group,and that of mild reaction in the 532 nm group was 33.3%.The
