摘 要:目的:研究九一丹对泡沫细胞(FM)内胆固醇逆转运及细胞凋亡的影响。方法:通过荧光标记氧化低密度脂蛋白诱导小鼠巨噬细胞RAW264.7形成FM。实验分对照组、模型组、九一丹组和Z-VAD-FMK组。采用荧光显微镜拍摄记录FM形成和细胞内脂质蓄积情况;油红O染色比较细胞内脂质沉积差异;生化试剂盒检测细胞内总胆固醇(TC)、游离胆固醇(FC)含量以及胆固醇流出率;TUNEL染色观察细胞凋亡;WesternBlot检测胆固醇逆转运相关蛋白,ATP结合盒转运子A1、G1(ABCA1、ABCG1)和清道夫受体-B1(SR-B1)的表达情况。结果:油红O染色和荧光显微镜下观察发现模型组细胞内脂质沉积增加,九一丹和Z-VAD-FMK处理后可有效降低FM内脂质;九一丹和Z-VAD-FMK干预后可降低TC、FC的水平,增加胆固醇流出率。与对照组比较,模型组ABCA1蛋白表达上调,SR-B1下调,九一丹和Z-VAD-FMK干预后ABCA1表达下调,SR-B1表达上调;TUNEL染色结果显示细胞凋亡较模型组减少。结论:九一丹能够上调胆固醇逆转运相关的SR-B1蛋白表达,促进细胞内脂质外排,减少FM凋亡发生,该过程为九一丹提脓祛腐表现提供了科学解释。Objective:To elucidate the impact of Jiuyi Dan on cholesterol reverse transport and apoptosis within foamy macrophages(FM).Methods:Mouse macrophage RAW264.7 cells were induced to form FM using Dil-oxidized low-density lipoprotein(Dil-Ox-LDL).The experimental design included control,model,Jiuyi Dan,and Z-VAD-FMK groups.Intracellular lipid accumulation and FM formation were visualized under fluorescence microscopy;lipid deposition was quantified using oil red O staining;total cholesterol(TC)and free cholesterol(FC)levels,as well as cholesterol efflux rates,were assessed with respective assay kits;apoptosis was evaluated by TUNEL staining;Western Blot analysis was conducted to determine the expression levels of ATP-binding cassette transporters A1 and G1(ABCA1,ABCGI)and scavenger receptor class B type 1(SR-B1),which are implicated in cholesterol reverse transport.Results:Oil red O staining and fuorescence microscopy observations revealed an increase in intracellular lipid deposition in the model group cells.Treatment with Jiuyidan and Z-VAD-FMK effectively reduced fluorescence intensity,suggesting a decrease in lipid content within FM.These treatments also decreased TC and FC levels and enhanced cholesterol efflux rates.Protein expression analysis revealed that the model group exhibited increased ABCA1 expression,and decreased SR-B1 expression compared to the control group.Following Jiuyi Dan and Z-VAD-FMK interventions,ABCA1 expression was downregulated,and SR-B1 expression was upregulated;TUNEL staining demonstrated a reduction in apoptotic cells.Conclusion:Jiuyi Dan upregulates the expression of the cholesterol reverse transport-related protein SR-B1,promotes the extrusion of intracellular lipids,and reduces the occurrence of FM apoptosis.This process provides a scientific explanation for the purulent abscess resolution phenotype.
