中药有效成分调控TGF-β信号通路干预心肌纤维化的研究进展

摘　　要：心肌纤维化(MF)是威胁人类健康的重要因素,预后较差。西医治疗主要以抗纤维化为主,但目前抗纤维化药物较少,且不良反应多,临床的运用具有局限性。而中医药在改善MF的病理改变及临床症状方面具有独特的优势。转化生长因子-β(TGF-β)信号通路是MF发生和发展的关键通路,参与胚胎发育、细胞增殖、肿瘤生长和细胞凋亡等各种生物过程。大量研究表明中药有效成分通过调控TGF-β信号通路,抑制胶原蛋白合成和降解、抗炎、抗氧化应激及减少细胞亡等多途径抑制细胞外基质沉积,进而保护心肌细胞及心脏功能。因此,文章着点于TGF-β信号通路及其与MF的关系,以及基于TGF-β通路干预MF的中药有效成分进行综述,旨在为中医药防治MF提供参考。Myocardial fibrosis(MF)is an important factor threatening human health and has a poor prognosis.Western medicine mainly focuses on anti-fibrosis,but at present,anti-fibrosis drugs are few and have great side effects,so the clinical application has limitations.Traditional Chinese medicine(TCM)has unique advantages in improving the pathological changes and clinical symptoms of MF.Transforming growth factor-β(TGF-β)signaling pathway is a key pathway in the pathogenesis and development of MF,which is involved in various biological processes such as embryonic development,cell proliferation,tumor growth and cell apoptosis.A large number of studies have shown that the effective components of TCM can inhibit extracellular matrix deposition by regulating TGF-βsignaling pathway,inhibiting collagen synthesis and degradation,anti-inflammatory,antioxidant stress and reducing cell apoptosis,and so on,so as to protect myocardial cells and cardiac function.Therefore,in this paper,TGF-βsignaling pathway and its relationship with MF,as well as the effective components of TCM based on TGF-βpathway intervention in MF were reviewed,aiming to provide reference for TCM prevention and treatment of MF.
