五子衍宗丸通过Rictor/mTORC2-PKC通路调控睾丸支持细胞骨架蛋白的机制研究

摘　　要：目的:研究五子衍宗丸含药血清对Rictor siRNA敲降后支持细胞骨架相关蛋白的影响。方法:使用CCK-8法筛选出五子衍宗丸含药血清的最大无毒浓度,然后采用siRNA技术降低支持细胞中Rictor的表达。五子衍宗丸含药血清干预后通过WesternBlot分析各组骨架相关蛋白表达,免疫荧光染色检测F-actin及β-tubulin的表达及定位。结果:五子衍宗丸含药血清可以促进p-PKC/PK表达(P<0.05);同时,五子衍宗丸含药血清干预后F-actin及β-tubulin平均光密度显著升高(P<0.05),同时F-actin及β-tubulin排列更为紧实、密集、荧光强度更强。结论:五子衍宗丸含药血清能改善Rictor沉默后支持细胞骨架相关蛋白表达,这可能与调节Rictor/mTORC2-PKC通路有关。Objective:To study the effects of drug-containing serum of Wuzi Yanzong Pills on cytoskeleton-related proteins after Rictor siRNA knockdown.Methods:The maximum non-toxic concentration of the serum containing Wuzi Yanzong Pills was screened by CCK-8 method,and then the expression of Rictor in supporting cells was decreased by siRNA technique.After intervention with the drug-containing serum of Wuzi Yanzong Pills,the expression of skeletal related proteins in each group was analyzed by Western Blot,and the expression and localization of F-actin andβ-tubulin were detected by immunofluorescence staining.Results:The serum containing Wuzi Yanzong Pills could promote the expression of P-PKC/PK(P<0.05).At the same time,the mean optical density of F-actin andβ-tubulin increased significantly after the intervention of drug-containing serum in Wuziyanzong Pills(P<0.05),while the arrangement of F-actin andβ-tubulin was more compact,dense,and the fluorescence intensity was stronger.Conclusion:The drug-containing serum of Wuzi Yanzong Pills can improve the expression of cytoskeletonrelated proteins after Rictor silencing,which may be related to the regulation of Rictor/mTORC2-PKC pathway.
