大黄[庶虫]虫丸干预肺纤维化小鼠最佳剂量研究

摘　　要：目的:探讨大黄[庶虫]虫丸不同剂量对博来霉素(BLM)致肺纤维小鼠的干预效果。方法:将C57BL/6小鼠分为6组,BLM造模后,以大黄虫丸高、中、低剂量灌胃给药,观察各组小鼠状态,绘制生存曲线、体质量变化曲线。取肺组织行HE和Masson染色,试剂盒检测羟脯氨酸(HYP),WesternBlot检测转化生长因子-β1(TCF-β1),IHC检测α-平滑肌肌动蛋白(α-SMA),评价各组小鼠肺纤维化程度。结果:6组小鼠中,除空白组外,死亡率、体质量下降程度、HYP含量以模型组更高,低剂量组更低;肺系数以模型组更高,吡非尼酮组更低;胶原纤维面积、TGF-β1、α-SMA含量模型组显著升高,低剂量组显著降低(P<0.05)。结论:大黄[庶虫]虫丸对BLM致肺纤维化小鼠确有一定的干预作用,且效果以低剂量最佳。Objective:To investigate the effects of different doses of Dahuang Zhechong Pills on the intervention of Bleomycin(BLM)-induced pulmonary fibrosis in mice.Methods:C57BL/6 mice were divided into 6 groups,and after BLM modeling,Dahuang Zhechong Pills was administered by gavage at high,medium and low doses to observe the status of the mice in each group,and to draw survival curves and curves of the changes in body mass,and the lung tissues were extracted to be stained by HE and Masson staining,HYP was detected by kit,TGF-β1 was detected by Western Blot,andα-SMA was detected by IHC,and the degree of pulmonary fibrosis in each group of mice was evaluated.Except for the blank group,the mortality rate,body mass downward potential and HYP content were higher in the model group and lower in the low-dose group,the lung coefficient was higher in the model group and lower in the pirfenidone group,and the collagen fiber area,TGF-β1,andα-SMA content were significantly elevated in the model group and significantly decreased in the low-dose group(P<0.05).Conclusion:Dahuang Zhechong Pills do have a certain intervention effect on BLM-induced pulmonary fibrosis mice,and the effect is best at low doses.
