摘 要:本研究采用超高效液相色谱-四极杆/静电场轨道高分辨质谱(UHPLC-Q-Exactive Orbitrap MS/MS)对草乌及其炮制品中的化学成分进行了快速鉴定。实验使用Acclaim 120 C_(18)色谱柱(2.1 mm×100 mm,3μm),以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,柱温为30℃,流速为0.3 mL·min^(-1),进样量为10μL。同时,基于电喷雾离子源(ESI),于正、负离子扫描模式下采集样品数据。通过数据解析,共鉴定得232个化合物,其中包括生物碱类105个(主要为C_(18)、C_(19)和C_(20)型二萜生物碱)、有机酸类54个、氨基酸及其衍生物33个、糖类11个、核苷酸9个和其他类20个。本研究结果能够为草乌及其炮制品药效物质基础研究提供参考,也可作为草乌药材深入研究的基础。This study used ultra-high performance liquid chromatography-quadrupole/electrostatic field orbitrap high-resolution mass spectrometry(UHPLC-Q-Exactive Orbitrap MS/MS) to rapidly identify the chemical components in Aconiti Kusnezoffii Radix and its processed products.The experiment used an Acclaim 120 C_(18) chromatographic column(2.1 mm×100 mm,3 μm) with acetonitrile 0.1%formic acid aqueous solution as the mobile phase for gradient elution.The column temperature was 30 ℃,the flow rate was 0.3 mL·min^(-1),and the injection volume was 10 μL.At the same time,based on the electric spray ion source(ESI),sample data are collected in the positive and negative ion scanning modes.Through data analysis,a total of 232 compounds were identified,including 105 alkaloids(mainly C_(18),C_(19),and C_(20) diterpenoid alkaloids),54 organic acids,33 amino acids and their derivatives,11sugars,9 nucleotides,and 20 other classes.The results of this study can provide reference for the research on the pharmacological substance basis of Aconiti Kusnezoffii Radix and its processed products,and can also serve as the basis for in-depth research on Aconiti Kusnezoffii Radix medicinal materials.
