摘  要:目的探究不同剂量槐杞黄颗粒对支气管哮喘(BA)模型小鼠气道重塑的影响及可能机制。方法本实验时间为2024年3—5月。选取SPF级雌性Balb/c小鼠50只,采用随机数字表法将其分为对照组、BA组、低剂量组、中等剂量组、高剂量组,每组10只。对照组小鼠不做任何处理,其他四组小鼠构建BA模型。低剂量组、中等剂量组、高剂量组小鼠从建模第15天开始给予槐杞黄颗粒灌胃,剂量分别为2、4、8 g/kg,BA组小鼠给予等体积0.9%氯化钠溶液灌胃,均连续灌胃28 d。采用乙酰甲胆碱(Mch)激发试验检测小鼠吸入6.25、12.50、25.00、50.00 g/L Mch时气道反应性〔采用增强呼气间歇(Penh)值评估〕,采用细胞计数仪检测小鼠肺泡灌洗液(BALF)中嗜酸粒细胞百分比,采用酶联免疫吸附试验检测小鼠BALF中炎症因子(IL-4、IL-5、IL-17水平),采用苏木精-伊红染色观察小鼠肺组织病理形态,采用TUNEL法检测小鼠肺上皮细胞凋亡率,采用Western blotting法检测小鼠肺组织IL-36、IL-36R、髓样分化因子88(MyD88)蛋白表达水平。结果BA组、低剂量组、中等剂量组、高剂量组小鼠吸入6.25、12.50、25.00、50.00 g/L Mch时Penh值、BALF中嗜酸粒细胞百分比及BALF中IL-4、IL-5、IL-17水平高于对照组,低剂量组、中等剂量组、高剂量组小鼠吸入6.25、12.50、25.00、50.00 g/L Mch时Penh值、BALF中嗜酸粒细胞百分比及BALF中IL-4、IL-5、IL-17水平低于BA组,中等剂量组、高剂量组小鼠吸入6.25、12.50、25.00、50.00 g/L Mch时Penh值、BALF中嗜酸粒细胞百分比及BALF中IL-4、IL-5、IL-17水平低于低剂量组,高剂量组小鼠吸入6.25、12.50、25.00、50.00 g/L Mch时Penh值、BALF中嗜酸粒细胞百分比及BALF中IL-4、IL-5、IL-17水平低于中等剂量组(P<0.05)。苏木精-伊红染色结果显示:BA组小鼠肺组织支气管炎症浸润严重,肺组织结构明显破坏,气道壁较厚;与BA组比较,低剂量组、中等剂量组、高剂�Objective To investigate the effects of different doses of Huaiqihuang granule on airway remodeling in bronchial asthma(BA)model mice and their potential mechanisms.Methods The experiment was conducted from March to May 2024.Fifty SPF-grade female Balb/c mice were selected and randomly divided into control group,BA group,low-dose group,medium-dose group,and high-dose group,with 10 mice in each group.The mice in the control group did not receive any treatment,and the other four groups of mice were constructed BA models.The mice in the low-dose group,the medium-dose group and the high-dose group were given Huaiqihuang granules by gavage from the 15 th day of modeling,and the doses were 2,4 and 8 g/kg,respectively.The mice in the BA group were given an equal volume of 0.9%sodium chloride solution by gavage,all rats were given intragastric administration for 28 d.Airway reactivity[assessed by enhance pause(Penh)]when the mice inhaled 6.25,12.50,25.00 and 50.00 g/L methacholine(Mch)was measured by Mch excitation test.The percentage of eosinophils in bronchoalveolar lavage fluid(BALF)of mice was detected by cell counter.The inflammatory factors(levels of IL-4,IL-5,and IL-17)in BALF of mice were detected by ELISA.Lung histopathology was assessed via hematoxylin-eosin staining.Apoptosis of lung epithelial cells was evaluated by TUNEL assay.Western blotting method was used to measure protein expression levels of IL-36,IL-36R,and myeloid differentiation factor 88(MyD88)in lung tissues.Results The values of Penh when mice inhaled 6.25,12.50,25.00 and 50.00 g/L Mch,the percentage of eosinophils in BALF and the levels of IL-4,IL-5 and IL-17 in BALF in BA group,low-dose group,medium-dose group and high-dose group were higher than those in control group,and all the above indicators in low-dose group,medium-dose group and high-dose group were lower than those in BA group,and all the above indicators in medium-dose group and high-dose group were lower than those in low-dose group,and all the above indicators in high-dose group we
