摘 要:目的分析黄芪总皂苷对实验性自身免疫性脑脊髓炎(encephalitis-autoimmune encephalitis,EAE)小鼠T细胞免疫调节的影响。方法2021年10月—2022年5月,于研究所动物实验中心选取24只C57BL/6雌性小鼠随机平均分为三组分别是正常对照组、EAE模型组、黄芪总皂苷治疗组,正常对照组未实施针对性处理措施,注射生理盐水,其他两组构建EAE模型,黄芪总皂苷治疗组每日300 mg/kg注射治疗。记录各组体质量、临床评分以及T细胞亚群百分比。结果给药14 d和28 d时,EAE组小鼠体质量较对照组显著下降,症状评分显著升高(P<0.05),黄芪总皂苷治疗组小鼠体质量丢失较EAE组减少,症状评分显著下降(P<0.05);给药28 d后,EAE模型组小鼠外周脾脏三个核细胞T细胞亚群百分比较正常对照组显著升高(P<0.05),黄芪总皂苷组INF-γ+CD4+T和IL-17+CD4+T细胞百分比较EAE组显著下降,IL-10+CD4+T和TGF-β+CD4+T细胞百分比较EAE组显著上升(P<0.05)。结论黄芪总皂苷能够促使EAE小鼠临床症状缓解,作用机制可能是其促进炎性T细胞数量下降和抑炎T细胞数量增加,对T细胞免疫调节产生显著影响。Objective To analyze the effects of total astragalus saponins on T cell immune regulation in mice with experimental Autoimmune encephalomyelitis(Autoimmune Encephalitis)(EAE).Methods From October 2021 to May 2022,24 female C57BL/6 mice were selected in the Animal experimental Center of the Institute and randomly divided into three groups.The normal control group was not treated with targeted treatment measures,and the other two groups were constructed with EAE model.The total Astragalus saponin treatment group was treated with 300mg/kg injection per day.Body mass,clinical score and percentage of T cell subsets were recorded in each group.Results On 14 and 28 days of administration,the body mass of mice in EAE group was significantly decreased compared with the control group,and the symptom score was significantly increased(P<0.05);the body mass loss of mice in total saponin group was reduced compared with the EAE group,and the symptom score was significantly decreased(P<0.05);After 28 days of administration,the percentage of T cell subsets of three nuclear cells in peripheral spleen of mice in EAE model group was significantly higher than that in normal control group(P<0.05),while the percentage of INF-γ+CD4+T and IL-17+CD4+T cells in Astragalus total saponins group was significantly lower than that in EAE group.The percentage of IL-10+CD4+T and TGF-β+CD4+T cells in EAE group were significantly increased(P<0.05).Conclusion Astragalus saponins can alleviate the clinical symptoms of EAE mice,and the mechanism of action may be that it promotes the decrease of the number of inflammatory T cells and the increase of the number of anti-inflammatory T cells,and has a significant effect on the immune regulation of T cells.
