摘 要:目的:通过16S rDNA测序对柴芍六君汤调节慢性萎缩性胃炎(CAG)胃黏膜屏障的作用机制进行探讨。方法:将36只Wistar大鼠按随机数字表法随机分为正常组8只和造模组28只,造模成功后将大鼠分为模型组、替普瑞酮组、柴芍六君汤组,每组8只,正常组和模型组大鼠给予灭菌饮用水灌胃,其余大鼠给予相应药物灌胃,持续4周。观察大鼠宏观指标、胃黏膜组织形态,检测大鼠血清白细胞介素-1β(IL-1β)、IL-18、IL-32、胃泌素-17(GAS-17)含量,胃黏膜组织一氧化氮(NO)、内皮型一氧化氮合成酶(eNos)、三叶因子2(TFF2)、前列腺素E_(2)(PGE_(2))mRNA表达水平,16S rDNA测序分析大鼠肠道菌群,并分析菌群和屏障保护因子的相关性。结果:与模型组比较,柴芍六君汤组和替普瑞酮组大鼠宏观指标、胃黏膜组织形态结果改善,IL-1β、IL-18、IL-32、GAS-17含量及PGE_(2)mRNA表达降低(P<0.01,P<0.001),NO、eNos、TFF2 mRNA表达升高(P<0.05,P<0.01)。替普瑞酮组异杆菌属、普雷沃菌属等和柴芍六君汤组乳酸杆菌属、帕拉普氏菌属等丰度降低(P<0.05,P<0.01,P<0.001)。乳酸杆菌属、异杆菌属等与NO、eNos、TFF2 mRNA水平负相关,与PGE_(2)mRNA水平正相关,脱硫弧菌属、阿氏菌属等与NO、eNos、TFF2 mRNA水平正相关,与PGE_(2)mRNA水平负相关(P<0.05)。结论:柴芍六君汤可能通过调节菌群丰度来恢复菌群的稳态,从而降低胃黏膜炎症反应,达到保护胃黏膜屏障的目的。Objective:To explore the mechanism by which Chaishao Liujun Decoction regulates the gastric mucosal barrier in chronic atrophic gastritis(CAG)through 16S rDNA sequencing.Methods:Thirty-six Wistar rats were randomly divided into a normal group(n=8)and a modeling group(n=28).After successful modeling,the rats were further divided into a model group,a teprenone group,and a Chaishao Liujun Decoction group,with eight rats in each group.The normal and model groups received sterile drinking water by gavage,while the other groups received their respective treatments for four weeks.Macroscopic indicators and gastric mucosal morphology were observed,and serum levels of interleukin-1β(IL-1β),IL-18,IL-32,and gastrin-17(GAS-17)were measured.The mRNA expression levels of nitric oxide(NO),endothelial nitric oxide synthase(eNOS),trefoil factor 2(TFF2),and prostaglandin E_(2)(PGE_(2))in gastric mucosal tissue were also analyzed.Additionally,16S rDNA sequencing was used to assess intestinal flora composition,and correlations between microbiota and barrier-protecting factors were examined.Results:Compared with the model group,the Chaishao Liujun Decoction and teprenone groups showed improvements in macroscopic indicators and gastric mucosal morphology,along with reduced levels of IL-1β,IL-18,IL-32,GAS-17,and PGE_(2)mRNA expression(P<0.01,P<0.001).Meanwhile,NO,eNOS,and TFF2 mRNA expression levels increased(P<0.05,P<0.01).The teprenone group exhibited a decrease in Aliikangiella and Prevotella abundance,while the Chaishao Liujun Decoction group showed a reduction in Lactobacillus and Paraprevotella abundance(P<0.05,P<0.01,P<0.001).Lactobacillus and Aliikangiella were negatively correlated with NO,eNOS,and TFF2 mRNA levels but positively correlated with PGE_(2)mRNA levels.Conversely,Desulfovibrio and Akkermansia were positively correlated with NO,eNOS,and TFF2 mRNA levels but negatively correlated with PGE_(2)mRNA levels(P<0.05).Conclusion:Chaishao Liujun Decoction may restore intestinal flora homeostasis by modulating microbial ab
