摘 要:旨在精准探究桂糖系列甘蔗品种试管苗在生根进程中相关激素的动态变化特征,深度剖析激素在甘蔗试管苗生根过程中的内在作用机制,进而为甘蔗试管苗生根培养方案的优化提供理论指引。以桂糖系列组培苗作为研究对象,测定了1~28天内生长素(IAA)、玉米素核苷(ZR)和脱落酸(ABA)的含量变化情况。研究结果表明,在完整的试验周期内,桂糖42号、桂糖47号和桂糖49号这3个品种的IAA含量整体呈现出逐步递减的趋势,ABA含量则显著上升,且均在第28天达到峰值。与此同时,在生根初期,桂糖42号、桂糖47号和桂糖49号的ZR含量保持相对稳定的小幅波动状态,然而随着生根过程的不断推进,ZR含量开始持续攀升。此外,本研究还发现,在整个试验阶段,上述3个品种的IAA/ABA比率普遍呈下降走向,IAA/ZR比值在整个试验周期内总体亦呈下降态势。研究成果为甘蔗组培生根条件的精准优化,如根据内源激素的变化情况补充外源激素,以及通过了解生根过程的激素变化与生根以及成活的关系,进而为前期移栽前激素处理的优化提供技术支撑。This study aims to precisely explore the dynamic change characteristics of relevant hormones in the rooting process of in vitro seedlings of the Guitang series sugarcane varieties,deeply analyze the internal action mechanism of hormones during the rooting process of sugarcane in vitro seedlings,and further provide a theoretical guidance for the optimization of the rooting culture scheme of sugarcane in vitro seedlings.Taking the tissue-cultured seedlings of the Guitang series as the research objects,the content changes of indole-3-acetic acid(IAA),zeatin riboside(ZR),and abscisic acid(ABA)were carefully determined during the three key stages,namely the rooting induction period(1~3 days),primordium formation period(4~7 days),and root system elongation period(8~28 days).The research results show that within the complete test cycle,the IAA content of the three varieties,Guitang42,Guitang47,and Guitang49,generally shows a gradually decreasing trend as a whole,while the ABA content significantly increases and reaches its peak value on the 28th day.At the same time,in the early stage of rooting,the ZR content of Guitang42,Guitang47,and Guitang49 remains in a state of relatively stable small fluctuations.However,with the continuous progress of the rooting process,the ZR content begins to rise continuously.In addition,this study also finds that during the entire test stage,the IAA/ABA ratio of the above three varieties generally shows a downward trend,and the IAA/ZR ratio also generally shows a downward trend within the entire test cycle.The findings of this study provide precise optimization for sugarcane tissue culture rooting conditions,such as supplementing exogenous hormones based on changes in endogenous hormone levels,and by understanding the hormonal changes during rooting and their relationship with root formation and survival,thereby providing technical support for the optimization of pre-transplantation hormone treatments.
