摘 要:基于免疫抑制动物模型分析山西恒山黄芪的根部、茎叶、花和蜂蜜的免疫调节作用。将SD大鼠随机分为6组(空白、模型、黄芪根、黄芪茎叶、黄芪花及黄芪蜂蜜),采用腹腔注射环磷酰胺(Cyclophos-phamide,CTX,40 mg/kg)诱导免疫抑制大鼠模型,造模3 d后,第4天给空白组和模型组灌胃蒸馏水,其余组灌胃黄芪不同部位提取物,连续灌胃10 d。试验结束后,取材计算大鼠脾脏、胸腺指数,苏木素-伊红染色(Hematoxylin-eosin staining,H&E)观察大鼠脾脏和胸腺的病理变化,免疫组化染色(Immunohisto-chemistry,IHC)分析黄芪不同部位对脾脏炎性因子的影响,酶联免疫吸附测定法(Enzyme-linked immuno-sorbent assay,ELISA)测定大鼠血清中免疫球蛋白G(Immunoglobulin G,IgG)、细胞因子干扰素-γ(Inter-feron-γ,IFN-γ)和肿瘤坏死因子α(Tumor necrosis factor-α,TNF-α)的含量。结果显示,模型组大鼠胸腺指数显著低于空白组(P<0.01),脾脏和胸腺组织呈现病理结构损伤,脾脏炎性因子表达降低,血清中TNF-α和IgG显著降低(P<0.05),IFN-γ也有一定程度的降低。与模型组比较,HQG组胸腺指数升高(P<0.05),脾脏和胸腺组织病理损伤减轻,HQY组TNF-α含量显著增加(P<0.05),黄芪不同部位提取物组脾脏IL-6、IL-1β表达水平呈现升高趋势。黄芪根、茎叶、花、蜂蜜均可以改善环磷酰胺诱导的免疫抑制,从而表现出不同程度的免疫调节作用。The immunomodulatory effects of roots,stems and leaves,flowers and honey of Shanxi Hengshan Astragalus were analyzed based on immunosuppressive animal models.SD rats were randomly divided into 6 groups(control,model,Astragalus root,Astragalus stem and leaf,Astragalus flower and Astragalus honey).The immunosuppressive rat model was induced by intraperitoneal injection of cyclophosphamide(CTX,40 mg/kg).After 3 days of modeling,the control group and the model group were given distilled water on the 4 th day,and the other groups were given different parts of Astragalus extract for 10 days.At the end of the experiment,the spleen and thymus indexes of rats were calculated.Hematoxylin-eosin staining(H&E)was used to observe the pathological changes of spleen and thymus in rats.Immunohistochemistry(IHC)was used to analyze the effects of different parts of Astragalus on inflammatory factors in spleen.Enzymelinked immunosorbent assay(ELISA)was used to determine the contents of Immunoglobulin G(IgG),Interferon-γ(IFN-γ)and Tumor necrosis factor-α(TNF-α)in rat serum.The thymus index of the model group was significantly lower than that of the control group(P<0.01).The pathological structure of the spleen and thymus tissue was damaged,the expression of inflammatory factors in the spleen was decreased,the serum TNF-α and IgG were significantly decreased(P<0.05),and IFN-γ was also decreased.Compared with the model group,the thymus index of HQG group was increased(P<0.05),the pathological damage of spleen and thymus tissue was alleviated,the content of TNF-α in HQY group was significantly increased(P<0.05),and the expression levels of IL-6 and IL-1β in spleen of different parts of Astragalus extract groups showed an increasing trend.The roots,stems and leaves,flowers and honey of Astragalus can improve the immunosuppression induced by cyclophosphamide,thus showing different degrees of immune regulation.
