血清半乳糖凝集素3对小鼠心肌纤维化的诊断价值

摘　　要：目的本实验分别应用低剂量和高剂量异丙肾上腺素(Isoproterenol,ISO)诱导ICR小鼠心肌纤维化,检测各组ICR小鼠半乳糖凝集素3(Galectin-3,Gal-3)血清水平,探讨血清半乳糖凝集素3对小鼠心肌纤维化的诊断价值。方法24只ICR小鼠随机数字表法分为3组,即对照组、异丙肾上腺素低剂量组和异丙肾上腺素高剂量组,每组8只。除对照组8只外,其余16只应用颈背部皮下注射ISO诱导心肌纤维化,异丙肾上腺素低剂量组皮下注射ISO 1 mg/(kg·d),异丙肾上腺素高剂量组皮下注射ISO_(2) mg/(kg·d),对照组给予同等剂量生理盐水,共4周。于实验4周末颈动脉采血后处死小鼠,留取小鼠心肌组织。称量体质量及心脏质量,计算心脏质量指数;HE染色;Masson染色;ELISA法测定各组ICR小鼠血清Galectin-3水平。结果异丙肾上腺素低剂量组和异丙肾上腺素高剂量组心脏质量指数高于对照组(P<0.001);HE染色与对照组相比,异丙肾上腺素低剂量组和异丙肾上腺素高剂量组心肌结构紊乱,高剂量组更加明显;Masson染色与对照组相比,异丙肾上腺素低剂量组和异丙肾上腺素高剂量组ICR小鼠心肌纤维化程度增强,并且可见心肌纤维断裂,高剂量组比低剂量组更明显;血清Galectin-3水平在异丙肾上腺素低剂量组和异丙肾上腺素高剂量组升高,高剂量组比低剂量组升高更加明显(P<0.001)。结论血清半乳糖凝集素3水平可反映小鼠心肌纤维化的程度,半乳糖凝集素3水平与心肌纤维化程度有剂量相关性。Objective This study applied different doses of isoproterenol to induce myocardial fibrosis in mice,detected serum galectin-3 levels,and explored the diagnostic value of Galectin-3 for myocardial fibrosis.Methods 24 ICR mice were randomly divided into three groups:control group,low-dose,and high-dose groups of isoproterenol.Except for the control group,which was given the same dose of normal saline,isoproterenol(1 mg/(kg·d),2 mg/(kg·d),respectively)was administered via subcutaneous injection for 4 weeks to induce myocardial fibrosis.Mice were euthanized at the end of the experiment to extract myocardial tissue.The following general indicators were measured:body mass,cardiac mass,and the cardiac mass index;HE staining,and Masson staining,were performed.ELISA was used to detect serum Galectin-3 levels in various groups of mice.Results The heart mass index in the isoproterenol groups were higher than those in the Control group(P<0.001).HE staining:Compared with the control group,the myocardial structure of the isoproterenol group was disordered;Masson staining:fiber fracture and vesicular degeneration were visible.The degree of fibrosis in the high-dose isoproterenol group was significantly higher than that in the low-dose isoproterenol group.The serum Galectin-3 level increased in the model group and was more pronounced in the high-dose group(P<0.001).Conclusion The level of serum Galectin-3 can reflect the degree of myocardial fibrosis in mice,and there is a dose-dependent relationship between Galectin-3 levels and the degree of myocardial fibrosis.
