摘 要:目的运用两样本孟德尔随机化方法探究体重指数与痛风之间的因果关系。方法暴露因素BMI的数据来源于欧洲IEU数据库(The IEU GWAS Database),疾病结局痛风的数据来源于芬兰数据库(The FinnGen Biobank),筛选出BMI相关的单核苷酸多态性位点,参数设为P<5×10^(-8),连锁不平衡r^(2)<0.001,kb<10000,孟德尔随机化分析主要运用IVW、ME、WME、SM和WM共5种互补的方法进行分析,以OR以及95%CI为依据评价BMI与痛风的之间因果关系。为确保研究结果的可靠性,还进行了敏感性分析,主要包括异质性检验、水平多效性、逐个剔除检验。结果IVW结果显示OR=1.627(95%CI:1.396~1.896),P=4.38E-10<0.01;MR-Egger结果显示OR=1.596(95%CI:1.063~2.397),P=0.025<0.05;WME结果显示OR=1.443(95%CI:1.218~1.708),P=2.17E-05<0.01;SM结果显示OR=1.194(95%CI:0.650~2.190),P=0.568>0.05;WM结果显示OR=1.383(95%CI:0.995~1.923),P=0.054>0.05。结论欧洲人群中随着体重指数的升高,罹患痛风的概率也会随之增加。Objective The two-sample Mendelian randomization method was used to explore the causal relationship between body mass index and Gout.Methods The data of exposure factor BMI were derived from The IEU GWAS Database,and the data of disease outcome Gout were derived from The FinnGen Biobank.SNPs associated with BMI were selected and the parameter was set as P<5×10^(-8),chain imbalance r^(2)<0.001,kb<10000,Mendelian randomization analysis mainly uses IVW,ME,WME,SM and WM,the causal relationship between BMI and gout was evaluated according to OR and 95%CI.In order to ensure the reliability of the research results,sensitivity analysis was also carried out,which mainly included heterogeneity test,horizontal pleiotropy test and one-by-one elimination test.Results IVW results showed that OR=1.627(95%CI:1.396~1.896),P=4.38E-10<0.01;The MR-Egger result showed that OR=1.596(95%CI:1.063~2.397),P=0.025<0.05;WME results showed that OR=1.443(95%CI:1.218~1.708),P=2.17E-05<0.01;SM results showed that OR=1.194(95%CI:0.650~2.190),P=0.568>0.05.WM results showed that OR=1.383(95%CI:0.995~1.923),P=0.054>0.05.Conclusion The risk of gout increases with the increase of body mass index in the European population.
