摘  要:线粒体铁蛋白(mitoferrin)与线粒体功能联系密切,而线粒体功能与帕金森病(Parkinson’s disease,PD)密切相关。为研究mitoferrin对帕金森疾病的影响,文章采用模式生物果蝇进行实验发现:鱼藤酮暴露下的PD模型果蝇寿命、运动能力受到显著影响,且PD模型果蝇的大脑活性氧(reactive oxygen species,ROS)荧光强度升高、腺苷三磷酸(ATP)水平降低;多巴胺能神经元mitoferrin OE转基因果蝇在鱼藤酮暴露下的寿命、运动能力、ROS积累等均有显著改善。结果表明,多巴胺能神经元mitoferrin OE可以通过恢复大脑线粒体产能降低大脑的ROS积累挽救鱼藤酮暴露导致的果蝇PD表型。该研究为探究PD的发病机制和治疗提供了一定的理论支持。Mitoferrin is closely associated with mitochondrial function,which is the focus in Parkinson’s disease(PD).To study the effect of mitoferrin on PD,this paper adopted model organism Drosophila melanogaster for experimentation.It was found that the life span and motor ability of wild Drosophila melanogaster were significantly affected by rotenone exposure,and the fluorescence intensity of reactive oxygen species(ROS)of Drosophila melanogaster brain increased,while the adenosine triphosphate(ATP)level decreased.However,the life span,motor ability,ROS accumulation and ATP level of Drosophila melanogaster with dopaminergic neuron mitoferrin overexpression(mitoferrin OE)under rotenone exposure were significantly rescued.The results show that the dopaminergic neuron mitoferrin OE may save the Drosophila melanogaster PD model caused by rotenone exposure by restoring the brain mitochondrial productivity and reducing the brain ROS accumulation.This study provides theoretical support for exploring the pathogeny and treatment of PD.
