玉米氮吸收性状相关NPF家族基因挖掘及InDel分子标记开发

摘  要:玉米作为中国的第一大粮食作物,在保障中国粮食安全方面的作用重大。在玉米生长发育过程中氮肥是重要的肥力因素,然而近些年氮肥的大量投入也造成了一系列环境问题。对于这些问题,利用分子标记辅助育种,充分发挥氮素的利用效率是一种环保高效的解决方法。本研究对硝态氮吸收转运相关的NPF家族进行分析,通过全基因组关联分析挖掘出与玉米氮吸收性状紧密关联的15个候选基因,根据已报道的功能位点设计插入缺失(InDel)分子标记,以期设计出与玉米硝酸盐含量积累性状显著关联的功能标记。共设计了69对InDel分子标记,经过亲本多态性检验,发现特异性良好的有32对标记。在609份玉米自交系组成的自然群体中对这些InDel分子标记进行功能验证,结果表明,在玉米ZmNPF6.2、ZmNPF6.4、ZmNPF6.8、ZmNPF8.12、ZmNPF8.14基因附近的7对InDel分子标记与氮素吸收性状紧密关联。本研究开发的氮高效吸收相关InDel分子标记可为分子标记辅助育种提供理论基础,为培育氮高效利用的玉米新品种指引了方向。As the largest grain crop in China,maize plays an important role in ensuring food security in our country.Nitrogen fertilizer is an important fertility factor in the growth and development of maize.However,in recent years,a series of environmental problems have been caused by a large amount of nitrogen fertilizer application.Molecular marker-assisted breeding which gives full play to nitrogen use efficiency is an environmentally friendly and efficient method for solving these problems.In this study,the NPF family related to nitrate nitrogen absorption and transport was analyzed,and 15 candidate genes closely associated with maize nitrogen absorption traits were dug out through genome-wide association analysis.Insertion-deletion(InDel)molecular markers were designed according to the reported functional sites,to design functional markers significantly associated with maize nitrate accumulation traits.A total of 69 pairs of InDel molecular markers were designed,and 32 pairs of markers with good specificity were tested by parental polymorphism.The functional validation of these InDel molecular markers in the natural population composed of 609 maize inbred lines showed that seven pairs of InDel molecular markers near the maize ZmNPF6.2,ZmNPF6.4,ZmNPF6.8,ZmNPF8.12 and ZmNPF8.14 genes were closely associated with nitrogen absorption traits.The InDel molecular markers related to efficient nitrogen absorption developed in this study can provide a theoretical basis for molecular marker-assisted breeding and guide the direction for breeding new maize varieties with efficient nitrogen utilization traits.
