摘 要:为探究以栅藻(Desmodesmus sp.CHX1)为藻种的藻菌共生体系的构建过程,通过因子优化的方法,探明了栅藻生长代谢适宜的氮源、碳源、pH和温度条件,分析了在最佳的生长因子组合下,废水原生微生物种群对栅藻接种的响应情况,并确定优势菌群,在优化条件下实现高效藻菌共生体系的构建。结果表明:促进栅藻生长和污染物降解的最佳组合为RS-6,即氨氮质量浓度为150 mg/L,葡萄糖和NaHCO_(3)的碳质量比为7∶3,温度为25℃,初始pH为6。此时总氮去除率为65.4%,总磷去除率为75.6%,氨氮去除率为80.8%,硝态氮去除率为71.6%,总叶绿素质量浓度达18.88 mg/L。栅藻的接种使废水微生物群落的物种丰富度迅速下降,高通量测序结果显示与栅藻共生的优势菌为施氏假单胞菌(Pseudomonas stutzeri)。In order to explore the construction process of symbiotic system with scenedesmus(Desmodesmus sp.CHX1)as algae species,the suitable nitrogen source,carbon source,pH and temperature conditions for scenedesmus growth and metabolism were found out by factor optimization method,and the response of wastewater protozoan population to scenedesmus inoculation was analyzed under the optimal combination of growth factors,and the dominant species was determined,so as to realize the construction of efficient symbiotic system of scenedesmus under the optimized conditions.The results showed that the optimal combination RS-6 for promoting the growth of scenedesmus and pollutant degradation was as follows:the mass concentration of ammonia nitrogen was 150 mg/L,the carbon mass ratio of glucose to NaHCO_(3)was 7∶3,the temperature was 25℃,and the initial pH was 6.Its total nitrogen removal rate was 65.4%,total phosphorus removal rate was 75.6%,ammonia nitrogen removal rate was 80.8%,nitrate nitrogen removal rate was 71.6%,and total chlorophyll mass concentration was up to 18.88 mg/L.Inoculation of scenedesmus rapidly reduced the species richness of microbial community in wastewater.High-throughput sequencing results showed that the dominant bacteria symbiotic with scenedesmus was Pseudomonas stutzeri.
