摘 要:目的研究排卵后老化(POA)对卵母细胞质量及子代行为学的影响,探讨卵母细胞的最佳受孕窗口期。方法雌性SD大鼠腹腔注射PMSG促进卵母细胞成熟,48 h后注射HCG促进卵母细胞排出后随机分组为排卵后AO 0 h组、AO 6 h组、AO 12 h组、AO 18 h组、AO 24 h组。每组取一部分雌性SD大鼠分离输卵管,获取卵母细胞,分别统计每组卵母细胞数量;体视显微镜观察卵母细胞形态及异常卵母细胞;采用Mito-Tracker Red CMXRos、JC-1和DCHF-DA荧光探针检测卵母细胞线粒体膜电位及活性氧(ROS)水平;另一部分雌性SD大鼠与雄鼠合笼受精到自然分娩,统计分析每组子鼠生长发育情况;Morris水迷宫检测每组子鼠学习记忆功能。结果随排卵时间延长,卵母细胞形态不规则、胞质间隙变宽、细胞破裂等异常率逐渐增加;ROS水平逐渐升高;线粒体膜电位下降。AO 12 h组分娩产子数量最多,但雄性子鼠组间生长体质量无差异,雌性子鼠生长体质量在3、6、8周时部分组别之间存在差异(P<0.05)。子鼠Morris水迷宫结果显示,AO 12 h组子鼠在逃离潜伏时间及距离上相对于其他组别占据优势,同时POA在一定程度上对子鼠学习记忆功能产生影响。结论自然状态下,雌鼠排卵12 h内为最佳受精窗期,可有效避免POA对受精率和子代造成的不良影响。Objective To study the effects of post ovulatory aging(POA)on the quality of oocytes and the behavior of offspring,and to explore the optimal window period for oocyte fertilization.Methods Female SD rats were intraperitoneally injected with pregnant mare serum gonadotropin(PMSG)to promote oocyte maturation.After 48 hours,HCG was injected to promote oocyte expulsion.Subsequently,the rats were randomly divided into the following groups:0-hour post ovulation(AO)group,AO 6-hour group,AO 12-hour group,AO 18-hour group,and AO 24-hour group.At the end of the experiment,the fallopian tubes of some female SD rats in each group were dissected to obtain oocytes,and the number of oocytes in each group was counted respectively;the morphology of oocytes and abnormal oocytes were observed under a stereomicroscope;Mito-Tracker Red CMXRos,JC-1 and DCHF-DA fluorescent probes were used to detect the mitochondrial membrane potential and reactive oxygen species(ROS)levels of oocytes.Another set of the female SD rats was caged with male rats for fertilization until natural delivery,and the growth and development of the offspring in each group were statistically analyzed;the morris water maze was used to detect the learning and memory functions of the adult offspring in each group.Results As the ovulation time prolonged,the abnormal rates of oocytes,such as irregular morphology,widened cytoplasmic gaps,and cell rupture,gradually increased;the ROS levels gradually rose;and the mitochondrial membrane potential decreased.The number of offspring born in the AO 12 h group was the largest,but there was no significant difference in growth weight between male rats groups.Female rats showed differences in growth weight between some groups at 3,6,and 8 weeks(P<0.05).The Morris water maze results of the 8-week-old offspring rats showed that the AO 12 h group had an advantage in escape latency and distance compared to other groups,and POA had a certain impact on the learning and memory function of the offspring rats.Conclusion Under natural condit
