摘  要:目的探讨贵州柞木原种提取液治疗高尿酸血症大鼠的药效机制。方法采用腹腔注射氧嗪酸钾和饲喂25%酵母膏饲料动物造模方法模拟HUA,并将60只大鼠分为空白对照组、模型组、模型+贵州柞木原种提取液高剂量组6.4 g/(kg·d)、中剂量组3.2 g/(kg·d)、低剂量组1.6 g/(kg·d)、模型+别嘌呤醇对照组75 mg/(kg·d)。连续灌胃3周后,测定各组大鼠尿尿酸含量及血尿酸含量、尿素氮、血肌酐水平,HE染色观察贵州柞木原种提取液对大鼠肾脏病理的影响,Western blot检测肝脏黄嘌呤氧化酶(XOD)蛋白、肾脏尿酸盐转运蛋白l(URAT1)蛋白的表达,并进行了贵州柞木原种提取液的主要化学成分及药毒检测。结果通过LC-MS技术对贵州柞木原种进行化学成分分析,筛选出含量排名前十的化合物,初步推测鞘脂类化合物二氢鞘氨醇及D-鞘氨醇可能与其降尿酸作用相关。贵州柞木原液与模型组比较,可明显降低模型大鼠尿尿酸水平,血尿酸水平,尿素氮水平,血肌酐水平(P<0.05)。HE显示贵州柞木原种提取液可改善炎症细胞浸润和肾小管萎缩,减小肾小球体积。Western blot显示贵州柞木原种提取液显著降低肝脏XOD蛋白、肾脏URAT1蛋白的表达(P<0.05)。结论贵州柞木原种提取液治疗高尿酸血症可能与改善肾功能、调节肝脏XOD蛋白和肾脏URAT1蛋白表达有关。Objective To explore the therapeutic mechanism of Guizhou xylosma japonicum original extract in treatment of hyperuricemia rats.Methods Simulation of hyperuricemia by means of animal modeling with intraperitoneal injection of potassium oxonate and feeding of 25%yeast paste diets,and rats were divided into blank control group,model group,model+Guizhou xylosma japonicum original extract high-dose group 6.4 g/(kg·d),medium-dose group 3.2 g/(kg·d),low-dose group 1.6 g/(kg·d)and allopurinol control group 75 mg/(kg·d).After 3 weeks of continuous gavage,the uric acid content in the urine and blood,urea nitrogen and serum creatinine levels of the rats in each group were measured.Hematoxylin-eosin(HE)staining was used to observe the effect of the extract of Guizhou xylosma japonicum original extract on the renal pathology of rats.Western blotting was employed to assess the expression levels of xanthine oxidase(XOD)protein in the liver and urate transporter 1(URAT1)protein in the kidney.The Guizhou xylosma japonicum original extract were also examined for their main chemical constituents.Results LC-MS was used to analyse the chemical constituents in the original species of Guizhou xylosma japonicum,and the top ten compounds were proposed,among which dihydrosphingosine and D-sphingosine might be related to the uric acid-lowering effect of the original species of Guizhou xylosma japonicum.Indicated that compared to the model group,the original extract of Guizhou xylosma japonicum significantly decreased urinary uric acid,blood uric acid,urea nitrogen,and blood creatinine levels in model rats(P<0.05);HE revealed that this extract improved kidney conditions by alleviating cyst luminal edema,renal tubular atrophy,and reducing glomerular volume.Additionally,Western blot analysis demonstrated that the extract significantly decreased the expression of hepatic XOD protein and kidney URAT1 protein(P<0.05).Conclusion The mechanism of Guizhou xylosma japonicum original extract in treating hyperuricemia may be related to improving
