摘 要:柱花草(Stylosanthes spp.)是全球热带地区栽培面积最大、应用最广泛的优良豆科牧草。由胶孢炭疽菌(Colletotrichum gloeosporioides)引起的柱花草炭疽病(Stylo anthracnose)是柱花草的一种毁灭性病害。炭疽菌转录因子CgHapX是铁稳态调控系统的关键因子,参与调控铁稳态途径相关基因的表达,与病原菌生长和致病力相关,目前有关柱花草胶孢炭疽菌CgHapX转录因子参与铁稳态调控系统相关研究尚未开展。本研究使用PCR克隆技术获得柱花草胶孢炭疽菌CgHapX基因cDNA全长序列,并通过生物信息学方法对序列进行结构和功能预测。结果表明:柱花草胶孢炭疽菌CgHapX基因包含一个1512 bp的开放阅读框架,编码504个氨基酸,其中脯氨酸含量最高,占比为11.70%;系统进化树显示,CgHapX蛋白与草莓炭疽病菌(C.aenigma,C.siamense)bZIP蛋白的亲缘关系为98.74%;该蛋白含典型bZIP结构域,二级结构主要由α-螺旋和无规则卷曲组成,分别占27.98%、71.43%,无跨膜结构,为亲水性蛋白。本研究为深入研究柱花草胶孢炭疽菌CgHapX基因分子生物学功能奠定理论基础,为靶标农药的生产及高效防控体系的建立提供理论依据。Stylosanthes spp.is the most widely cultivated and utilized leguminous forage grass in tropical regions worldwide,Stylo anthracnose,caused by Colletotrichum gloeosporioides,is a destructive disease of Stylosanthes.Transcription factor CgHapX of Colletotrichum gloeosporioides is key factor in iron homeostasis regulatory system,and is involved in the regulation of iron homeostasis pathogenicity of Colletotrichum gloeosporioides,which is related to the growth and pathogenicity of the pathogen.Currently,there are no studies on the role of the CgHapX transcription factor in the iron homeostasis regulatory system of C.gloeosporioides in Stylosanthes.In this study,the full-length cDNA sequence of CgHapX was cloned by PCR method,and the structure and function were predicted by bioinformatics methods.The results showed that CgHapX gene contained an 1512bp open reading frame,encoding 504 amino acids,with the highest content of proline reaching 11.70%.Systematic evolution analysis showed that CgHapX protein had the closest genetic 98.74% relationship with bZIP proteins of strawberry Colletotrichum(C.aenigma,C.siamense).This protein contained typical bZIP domain,and was a hydrophilic protein without a transmembrane structure,Alpha helix and random coil were the main components of the secondary structure of CgHapX protein,accounting for 27.98% and 71.43% respectively.There was no transmembrane structure,and it was a hydrophilic protein.This study lays a theoretical foundation for further research on the molecular biological function of the CgHapX gene,and provides a basis for the production of target pesticides and the establishment of an efficient prevention and control system.
