摘 要:目的 研究丹玉通脉颗粒对血管紧张素Ⅱ诱导的大鼠乳鼠心肌成纤维细胞活化、增殖和迁移的影响及其机制。方法 将分离成功的心肌成纤维细胞分为5组:对照组、模型组、丹玉通脉颗粒低剂量组(0.5 g·L^(-1))、丹玉通脉颗粒中剂量组(1 g·L^(-1))和丹玉通脉颗粒高剂量组(2 g·L^(-1))。对照组未接受血管紧张素Ⅱ和丹玉通脉颗粒,模型组接受0.1μmol的血管紧张素Ⅱ,加药组接受不同浓度丹玉通脉颗粒预处理心肌成纤维细胞2 h后,再与含有0.1μmol的血管紧张素Ⅱ培养基共孵育24 h。免疫荧光实验检测心肌成纤维细胞活化,MTT实验检测心肌成纤维细胞异常增殖,划痕试验检测心肌成纤维细胞迁移,Western blot实验检测相关通路蛋白表达。结果 和对照组相比,模型组心肌成纤维细胞增加,α-SMA蛋白表达增加(P<0.05),表明体外模拟心肌纤维化模型构建成功。和模型组相比,低、中和高剂量丹玉通脉颗粒组的细胞生存率和迁移率降低(P<0.05),细胞中p-P65蛋白表达量降低(P<0.05),并且随着丹玉通脉颗粒浓度的增加,抑制作用更明显。结论 丹玉通脉颗粒能够有效地抑制血管紧张素Ⅱ诱导的大鼠乳鼠心肌成纤维细胞活化、增殖和迁移,且抑制作用呈现浓度依赖趋势,其机制可能与抑制炎症通路(NF-κB)相关蛋白的表达有关。Objective To study the effects of Danyu Tongmai Granules on activation,proliferation and migration of cardiac fibroblasts induced by angiotensinⅡ.Methods The isolated neonatal rat myocardial fibroblasts were divided into 5 groups,namely control group,model group,low dose Danyu Tongmai Granules(0.5 g·L^(-1))group,medium dose Danyu Tongmai Granules(1 g·L^(-1))group and high dose Danyu Tongmai Granules(2 g·L^(-1))group.Control group was given normal medium,model group was given 1μmol angiotensinⅡ,and drug group was given different concentrations of Danyu Tongmai Granules to pretreat myocardial fibroblasts for 2 hours,then incubated with 1μmol angiotensinⅡfor 24 hours.MTT method was detected proliferation of myocardial fibroblasts.Migration was detected by scratch and Transwell method.Western blot method was used to detect the expression of myocardial fibroblast activation and collagen protein.Results Compared with control group,myocardial fibroblasts and collagen expression in model group increased(P<0.05),indicating that the in vitro myocardial fibrosis model was successfully constructed.Compared with model group,survival and migration rate of cell in drug group were significantly decreased(P<0.05),and collagen expression was decreased(P<0.05),Danyu Tongmai Granules concentration increased,inhibitory effect was more obvious.Conclusion Danyu Tongmai Granules can effectively inhibit activation,proliferation and migration of neonatal rat myocardial fibroblasts induced by angiotensinⅡ,and reduce the expression of collagen protein,and the inhibitory effect shows a concentration-dependent trend,the mechanism may be related with the inflammatory pathway(NF-κB).
