摘 要:目的探究甲基莲心碱对糖尿病肾病大鼠肾脏氧化应激的影响。方法将SD雄性大鼠随机分为对照组、模型组、治疗组和阳性对照组。除对照组外,其余三组予以高糖高脂饲料喂养制造DN模型。造模成功以后分组药物治疗。对照组和模型组使用等量盐水灌胃,治疗组使用40 mg/kg甲基莲心碱灌胃,阳性对照组使用厄贝沙坦14 mg/kg灌胃。HE染色光学显微镜下观察大鼠肾脏组织病理情况,生化分析大鼠尿液尿蛋白含量,ELISA检测大鼠肾脏组织TNF-α、SOD和MDA含量。结果与对照组相比,模型组肾脏组织结构受损明显,大鼠尿液尿蛋白显著增加,肾组织SOD含量显著下降,MDA和TNF-α含量显著上升(P<0.01);与模型组相比,治疗组局部肾脏组织不同程度受损,治疗组尿蛋白显著减少(P<0.01),SOD含量上升(P<0.05),MDA和TNF-α含量下降(P<0.05)。结论甲基莲心碱可改善糖尿病肾病大鼠肾脏组织损伤,降低尿蛋白含量,缓解氧化应激水平和炎症反应。Objective To investigate the effect of neferine on renal oxidative stress in rats with diabetic nephropathy.Methods SD male rats were randomly divided into control group,model group,treatment group and positive control group.In addition to the control group,the other three groups were fed with high-sugar and high-fat diet to make DN model.After successful modeling,grouped drug treatment.The control group and the model group were intragastrically administered with the same amount of saline,the treatment group was intragastrically administered with 40 mg/kg neferine,and the positive control group was intragastrically administered with 14 mg/kg irbesartan.HE staining was used to observe the pathological conditions of rat kidney tissue under optical microscope.Biochemical analysis of urinary protein content in rats was performed.ELISA was used to detect the contents of TNF-α,SOD and MDA in rat kidney tissue.Results Compared with the control group,the renal tissue structure of the model group was significantly damaged,the urine protein of rats was significantly increased,the content of SOD in renal tissue was significantly decreased,and the contents of MDA and TNF-αwere significantly increased(P<0.01).Compared with the model group,the local renal tissue of the treatment group was damaged to varying degrees,the urine protein of the treatment group was significantly reduced(P<0.01),the content of SOD was increased(P<0.05),and the content of MDA and TNF-αwas decreased in the treatment group(P<0.05).Conclusion Neferine can improve the damage of renal tissue in diabetic nephropathy rats,reduce the content of urinary protein,and alleviate the oxidative stress level and inflammatory response in the kidney group of diabetic nephropathy rats.
