摘 要:基于醛类化合物可与2,4-二硝基苯肼(DNPH)反应生成红色显色物质的原理,采集有颜色部位的图像,利用三基色红绿蓝(RGB)模式提取数据,建立了一种检测醛类化合物的数字图像比色法,用于检测肉桂油样品中醛类化合物,并对检测方法进行了条件优化和方法学验证。研究结果表明:针对20μL待测样品,加入1 mL的DNPH(5 mmol/L)、 10μL盐酸和1.5 mL氢氧化钾(60 g/L)完成显色反应,可利用样本颜色的蓝/绿(B/G)值作为指标建立肉桂醛测量方法,该方法标准曲线的相关系数为0.998 6,定量限为1 mg/L,检测限为0.5 mg/L,基质干扰为6.5%,日内和日间精密度分别为0.39%和0.52%,准确度为102.5%~107.5%,相对标准偏差(RSD)均小于6.5%。Based on the principle that aldehydes could react with 2,4-dinitrophenylhydrazine(DNPH)to produce red chromogenic substances,the images of colored parts were collected and the three-color red,green and blue(RGB)mode was used to obtain data.A digital image colorimetry method was established to detect aldehydes and applied to cinnamon oil samples,and the conditions of detection method were optimized and the methodology was verified.The results showed that for 20μL samples to be tested,1 mL DNPH(5 mmol/L),10μL hydrochloric acid and 1.5 mL potassium hydroxide(60 g/L)were added to complete the color reaction,and the cinnamaldehyde measurement method could be established using the blue/green(B/G)value of the sample color as an indicator.The correlation coefficient of the standard curve of the method was 0.9986,the limit of quantitation was 1 mg/L,the detection limit was 0.5 mg/L,the matrix interference was 6.5%,the intraday and daytime precision were 0.39%and 0.52%,respectively,the accuracy was 102.5%-107.5%,and the relative standard deviation(RSD)was less than 6.5%.
