摘  要:目的本研究旨在探讨FGL2对EBV感染的T细胞免疫应答的影响,包括T细胞的活化、增殖、耗竭状态以及细胞因子谱的变化。方法使用不同感染复数的EBV感染原代T淋巴细胞,通过RT-qPCR、Western blot、ELISA、CCK8以及流式细胞术等方法,检测FGL2在T细胞中的表达及T细胞的活化、增殖、耗竭和细胞因子水平。进一步通过敲低和过表达FGL2,观察FGL2对EBV感染的T细胞免疫应答的具体影响。结果FGL2在EBV感染的T细胞中表达显著上调(P<0.05)。同时,EBV感染后T细胞出现活化(P<0.001)、增殖(P<0.001)和耗竭(P<0.01)水平升高。与T细胞+EBV组相比,T细胞+EBV+FGL2过表达组T细胞的耗竭水平更高(P<0.01),活化(P<0.05)和增殖(P<0.05)程度更低,促炎性细胞因子水平降低(P<0.05),抑炎性细胞因子水平升高(P<0.05);而T细胞+EBV+FGL2敲低组则表现出相反的趋势,即T细胞的活化(P<0.01)、增殖水平更高(P<0.05),耗竭程度更低(P<0.01),促炎性细胞因子水平升高(P<0.05),抑炎性细胞因子水平降低(P<0.05)。结论FGL2在EBV感染中能抑制T细胞的活化和增殖,增加T细胞的耗竭程度,同时抑制促炎性细胞因子的释放并促进抑炎性细胞因子的释放,从而调控T细胞的免疫应答。Objective This study aims to investigate the effects of FGL2 on the immune response of EBV-infected T cells,including their activation,proliferation,exhaustion,and cytokine profile changes.Methods Primary T cells were infected with EBV at different multiplicities of infection(MOI).Expression of FGL2 in T cells,as well as T-cell activation,proliferation,exhaustion,and cytokine levels,were detected using RT-qPCR,Western blot,ELISA,CCK8,and flow cytometry(FCM),respectively.Further experiments involving FGL2 knockdown and overexpression were conducted to elucidate its specific regulatory role in EBV-infected T cells.Results FGL2 expression was significantly upregulated in EBV-infected T cells(P<0.05).EBV infection also induced enhanced T cell activation(P<0.001),proliferation(P<0.001),and exhaustion(P<0.01).Compared to the T cells+EBV group,the T cells+EBV+FGL2 overexpression group exhibited higher exhaustion levels(P<0.01),reduced activation(P<0.05)and proliferation(P<0.05),decreased pro-inflammatory cytokine levels(P<0.05),and increased anti-inflammatory cytokine levels(P<0.05).Conversely,the T cells+EBV+FGL2 knockdown group demonstrated the opposite trends,with elevated activation(P<0.01),proliferation(P<0.05),pro-inflammatory cytokine levels(P<0.05),and reduced exhaustion(P<0.01)and antiinflammatory cytokine levels(P<0.05).Conclusion FGL2 suppresses T cell activation and proliferation,exacerbates T cell exhaustion,inhibits pro-inflammatory cytokine release,and promotes anti-inflammatory cytokine secretion during EBV infection,thereby modulating the immune response of T cells.
