紫外分光光度法测定山楂提取物中的总黄酮含量

摘  要:以芦丁为标准品,经过三氯化铝显色,利用全波长紫外分光光度计扫描,并选择最大吸收波长为检测波长。利用超声辅助提取对山楂进行提取,显色后在最大吸收波长处检测黄酮含量,并进行方法学考察。结果显示:芦丁经三氯化铝显色后,紫外最大吸收波长为273nm;以芦丁质量浓度为横坐标,吸光度为纵坐标,建立芦丁标准曲线为y=0.0348x+0.0068,R^(2)=1;经计算样品山楂提取物中黄酮含量为15.49mg/g,RSD为1.77%;加标回收率范围为92.85%~95.76%。该方法简单可靠,稳定,适合山楂中黄酮含量的测定。Using rutin as the standard,the color was developed with aluminum trichloride,scanned with a full wavelength ultraviolet spectrophotometer,and the maximum absorption wavelength was selected as the detection wavelength.Using ultrasound assisted extraction to extract hawthorn,detecting flavonoid content at the maximum absorption wavelength after color development,and conducting methodological investigations.The results showed that the maximum UV absorption wavelength of rutin after color development with aluminum trichloride was 273 nm;A standard curve for rutin was established with the mass concentration of rutin as the x-axis and absorbance as the yaxis,with y=0.0348x+0.0068,R^(2)=1;After calculation,the flavonoid content in the hawthorn extract sample was 15.49 mg/g,with an RSD value of 1.77%;The recovery rate of spiked samples ranged from 92.85% to 95.76%.This method is simple,reliable,stable,and suitable for the determination of flavonoid content in hawthorn.
