摘 要:试验旨在探究黑果枸杞花青素提取物(anthocyanin extract of Lycium ruthenicum Murr,AEL)通过调控八眉三元猪血常规、血气参数及Ca^(2+)稳态途径对心脏的保护作用。选择初始体重为(7.82±0.59) kg的八眉三元猪48头,随机分为4组,对照组(CON组)饲喂基础日粮,试验组在基础日粮中分别添加5(L-AEL组)、10(M-AEL组)、15 g/kg(H-AEL组)的AEL,饲养35 d。试验结束后分析各组血液学参数变化,利用qRT-PCR、Western blot检测Ca^(2+)稳态途径相关因子基因转录和蛋白表达水平。结果显示:(1)与CON组相比,添加AEL能够显著提高血常规参数红细胞数(RBC)、血红蛋白浓度(Hb)、红细胞压积(HCT)、平均血红蛋白浓度(MCHC)、平均血红蛋白量(MCH),显著降低血小板(PLT)、血小板压积(PCT)、平均血小板体积(MPV)(P<0.05);显著提高血气参数酸碱度(pH)、血氧分压(PO_(2))、血氧饱和度(SO_(2))、Na^(+)含量,显著降低K^(+)、Ca^(2+)、二氧化碳总量(TCO_(2))、阴离子间隙(AG)(P<0.05);(2)与CON组相比,添加AEL能够显著上调肌浆网钙ATP酶(SERCA)活性,显著降低猪心肌组织Ca^(2+)水平(P<0.05),显著下调钙敏感受体(CASR)、钙调蛋白(CaM)的基因转录和蛋白表达水平,显著上调质膜Ca^(2+)-ATP酶(PMCA)、SERCA的基因转录和蛋白表达水平(P<0.05)。综上所述,AEL通过调节八眉三元猪血常规及血气参数,提高其机体在低氧环境的携氧运输能力,有效改善高海拔低氧环境引起的八眉三元猪心肌组织中Ca^(2+)稳态失衡,发挥心脏保护作用,且添加量为10 g/kg时效果显著。The aim of this experiment was to investigate the cardioprotective effects of anthocyanin extract of Lycium ruthenicum Murr(AEL)by modulating blood routine,blood gas and Ca^(2+)homeostatic pathways in Bamei ternary porcine.Forty-eight Bamei ternary porcines with an initial weight of(7.82±0.59)kg were selected for the experiment and randomly divided into four groups.The control group(CON group)was fed a basal diet,and the experimental groups added 5 g/kg(L-AEL group),10 g/kg(M-AEL group),and 15 g/kg(H-AEL group)of AEL to the basal diet,respec-tively,and kept for 35 days.At the end of the experiment,we analyzed the changes of hemato-logical parameters in each experimental group,and detected the gene transcription and protein expression levels of factors related to Ca^(2+)homeo-static pathway using qRT-PCR and Western blot.The results showed that:①the effects of AEL on hematological parameters of Bamei ternary porcines,compared with the CON group,different additive amounts of AEL could significantly increase the hematological parameters red blood cell count(RBC),hemo-globin concentration(Hb),hematokrit(HCT),mean hemoglobin concentration(MCHC),mean hemoglobin volume(MCH),and significantly decrease platelets(PLT),platelet pressure product(PCT),mean platelet volume(MPV)(P<0.05);significantly increased blood gas parameters pH,partial pressure of oxygen(PO_(2)),oxygen saturation(SO_(2)),and Na^(+)content,and significantly decreased K^(+),Ca^(2+),total carbon dioxide(TCO_(2)),and anion gap(AG)(P<0.05).②Effect of AEL on Ca^(2+)homeostasis in heart tissues of Bamei ternary por-cines,compared with the CON group,different additions of AEL were able to significantly up-regulate sarco-plasmic reticulum calcium-ATPase(SERCA)activity,significantly reduce the Ca^(2+)level in porcine myocar-dial tissues(P<0.05),significantly down-regulate the gene transcription and protein expression levels of calcium-sensitive receptor(CASR),calmodulin(CaM),and significantly up-regulate the gene transcription and protein expression levels of
