摘  要:研究以从新鲜驴粪中筛选出高产纤维素酶的枯草芽孢杆菌LV2为研究对象,优化其发酵产纤维素酶工艺。以单因素试验、响应面试验分析培养基营养成分(氮源、碳源、无机盐)和发酵条件(接种量、温度、时间、pH、转速)对纤维素酶活性的影响。结果显示,枯草芽孢杆菌LV2产纤维素酶的最佳发酵参数为:蔗糖54.4 g/L,酵母粉20.6 g/L,磷酸氢二钠5.2 g/L,接种量6%,培养温度37℃,培养时间24 h,发酵初始pH 7.0,转速200 r/min。在最佳发酵参数下,LV2经24 h发酵纤维素酶产量为59.19 U/mL,是优化前的3.08倍;枯草芽孢杆菌LV2在纤维素分泌和水解方面具有良好的应用前景。This study selected Bacillus subtilis LV2 with high cellulase production from fresh donkey dung as the research object,and optimized its fermentation technology for cellulase production.The effects of nutrient components of medium(nitrogen source,carbon source,inorganic salt)and fermentation conditions(inocula-tion amount,temperature,time,pH and rotational speed)on cellulase activity were analyzed by singlefactor testand response surface test.The results showed that the optimum fermentation parameters for cellulase pro-duction by Bacillus strains LV2 were as follows:sucrose 54.4 g/L,yeast powder 20.6 g/L,disodium hydrogen phosphate 5.2 g/L,inoculation amount 6%,culture temperature 37℃,culture time 24 h,initial pH 7.0,and optimal rotational speed 200 r/min.Under the opti-mal fermentation parameters,the cellulase yield of LV2 after 24 h fermentation was 59.19 U/mL,which was 3.08 times higher than that before opti-mization.Bacillus subtilis LV2 had a good applica-tion prospect in cellulose secretion and hydrolysis.
