摘 要:目的:考察淫羊藿的最佳提取工艺。方法:通过回流、超声和破壁法的对比研究,最终选取破壁法提取;在单因素试验基础上,以破壁时间、乙醇体积分数及液料比为影响因素,采用高效液相色谱法测定朝藿定A_1、朝藿定A、朝藿定B、朝藿定C和淫羊藿苷含量,计算指标总质量分数,并将其作为淫羊藿提取的评价指标,考察Box-Behnken响应面法优化的提取工艺;用显微镜观察回流、超声和破壁法提取所得提取液的状态。结果:破壁法提取淫羊藿的工艺最佳,破壁时间单因素试验中淫羊藿指标总质量分数最大,为26.54 mg·g^(-1),与回流法和超声法相比,分别高出约28.46%和21.80%;Box-Behnken响应面法优化最佳提取条件为破壁时间4 min,乙醇体积分数50%,液料比30∶1。在此条件下,淫羊藿指标总质量分数平均值为32.68 mg·g^(-1),总转移率平均值为94.72%,RSD为1.59%,优化的工艺稳定可行。显微镜观察表明,采用破壁法提取,淫羊藿叶片的组织能更充分接触溶剂,提取效率提高。结论:破壁法提取淫羊藿的效率更高,节约时间和能源,提高了转移率,对淫羊藿的提取生产工艺有指导作用。Objective:To investigate the optimal extraction technology of Epimedii Folium.Methods:The cell wall disruption method was selected for extraction by comparing it with the reflux extraction and ultrasonic-assisted extraction.Based on the single-factor experiment,the cell wall disruption time,ethanol concentration,and solid-to-liquid ratio were selected as the influencing factors.The high-performance liquid chromatography(HPLC)was used to determine the contents of epimedin A1,epimedin A,epimedin B,epimedin C,and icariin.The total extraction amount was used as the evaluation index.The Box-Behnken response surface method was used to optimize the extraction technology.The state of the liquid extracted by the reflux method,ultrasonic method,and cell wall disruption method was observed using a microscope.Results:The cell wall disruption method had the best extraction effect on Epimedii Folium;in a single-factor experiment of cell wall disruption time,the maximum total extraction amount was 26.54 mg·g^(-1),which was approximately 28.46%and 21.80%higher than that of the reflux method and ultrasound method,respectively.The optimal extraction conditions for Box-Behnken response surface method optimization were:cell wall disruption time of 4 min,ethanol concentration of 50%,and solid-to-liquid ratio of 30:1.Under these conditions,the total extraction amount was 32.68 mg·g^(-1);the total transfer rate was 94.72%,and the RSD value was 1.59%.The optimized process was stable and feasible.The microscopic observations showed that the tissue of Epimedii Folium leaves was more fully exposed to solvent,and the extraction efficiency was improved by using the cell wall disruption method.Conclusion:The cell wall disruption method had a higher efficiency for extracting Epimedii Folium.It could save time and energy and improve the transfer rate,which has a guiding effect on the extraction and production process of Epimedii Folium.
