摘 要:目的:探究新型冠状病毒对肺栓塞大鼠肺组织病理改变、血管紧张素转化酶2(ACE2)及血管紧张素Ⅱ(AngⅡ)表达水平的影响。方法:将60只大鼠随机分为四组:空白对照(C)组、肺栓塞模型(M)组、新型冠状病毒S蛋白(S)组及肺栓塞模型+新型冠状病毒S蛋白(MS)组,每组15只。利用明胶海绵建立急性肺栓塞模型,随后通过腹腔注射给予新型冠状病毒S蛋白或等量0.9%氯化钠注射液。分别于注射后第1、5和10天,收集肺组织进行肺病理学检查,评估肺损伤程度;采用实时荧光定量聚合酶链反应(RT–qPCR)技术检测ACE2信使核糖核酸(mRNA)和AngⅡmRNA的表达水平,通过酶联免疫吸附法(ELISA)方法检测血清中ACE2的含量及肺组织ACE2、AngⅡ蛋白的表达水平。结果:M组和S组大鼠的肺损伤症状随时间逐渐缓解,血清ACE2水平、肺组织ACE2 mRNA及ACE2蛋白的表达均呈现下调趋势,并逐渐恢复至正常水平;而MS组大鼠的肺损伤症状持续加重,血清ACE2水平长时间维持高水平状态,且肺组织中AngⅡmRNA和AngⅡ蛋白的表达显著升高。结论:肺栓塞后长时间高水平的血清ACE2及肺部AngⅡ蛋白的高表达是新型冠状病毒导致肺组织严重损伤的重要机制。Objective To investigate the effects of novel coronavirus on the pathological changes of lung tissue,angiotensin-converting enzyme 2(ACE2)and angiotensin II(AngⅡ)expression in rats with pulmonary embolism(PE).Methods Sixty rats were randomLy divided into four groups:blank control group(group C),PE model group(group M),novel coronavirus S protein group(group S)and PE+novel coronavirus S protein group(group MS),with 15 rats in each group.An acute PE model was established using gelatin sponge,followed by intraperitoneal injection of novel coronavirus S protein or an equivalent 0.9%sodium chloride injection.Lung tissue was collected at 1,5 and 10 days after injection for pulmonary examination to assess the degree of lung injury.Real-time fluorescence quantitative polymerase chain reaction(RT-QPCR)was used to detect the expression levels of ACE2 messenger RNA(mRNA)and AngⅡmRNA.The serum ACE2 content and lung ACE2 and AngⅡprotein expression levels were detected by enzyme-linked immunosorbination(ELISA).Results The lung injury symptoms of rats in group M and group S were gradually relieved with time,and the serum ACE2 level,the expression of lung ACE2 mrna and ACE2 protein showed a downward trend,and gradually returned to normal level.In the MS group,the symptoms of lung injury continued to worsen,the serum ACE2 level remained high for a long time,and the expression of AngⅡmRNA and AngⅡprotein in lung tissue was significantly increased.Conclusion The long-term high level of serum ACE2 and lung AngⅡprotein expression after PE is an important mechanism of severe lung tissue injury caused by novel coronavirus.
