摘　　要：目的检测重楼乙醇提取物对蝮蛇毒中主要酶活力的抑制作用,分析重楼乙醇提取物治疗蝮蛇咬伤的效果。方法通过平板实验检测重楼乙醇提取物对蝮蛇毒蛋白水解酶和磷脂酶A2的抑制作用,利用分光光度法检测重楼乙醇提取物对蝮蛇毒透明质酸酶活性的抑制作用,并通过血凝实验来评估重楼乙醇提取物对蝮蛇毒凝血酶活性的抑制作用。结果蝮蛇毒与重楼乙醇提取物的质量比达到1∶50时,重楼乙醇提取物能显著抑制蝮蛇毒中的磷脂酶A2和蛋白水解酶活性。当蝮蛇毒与重楼乙醇提取物的质量比达到1∶50时,蝮蛇毒透明质酸酶的活性可以被重楼乙醇提取物完全抑制。此外,重楼乙醇提取物可以显著抑制蝮蛇毒凝血作用。结论重楼乙醇提取物能够有效抑制蝮蛇毒中主要酶类的活性,为治疗蝮蛇咬伤提供一定的理论依据。Objective To investigate the inhibitory effect of Paris polyphylla ethanol extract on the main enzymatic activities in the Agkistrodon venom and explore the therapeutic efficacy of Paris polyphylla ethanol extract on the Agkistrodon snakebites.Methods The inhibitory effects of Paris polyphylla ethanol extract on Agkistrodon snake venom proteases and phospholipase A2 were detected through plate experiments.The inhibitory effect of Paris polyphylla ethanol extract on the activity of Agkistrodon venom hyaluronidase was detected using spectrophotometry,and the inhibitory effect on the coagulation enzyme activity of Agkistrodon venom was assessed through hemagglutination experiments.Results When the mass ratio of Agkistrodon venom to Paris polyphylla ethanol extract reached 1:50,the extract inhibited significantly the activities of phospholipase A2(PLA2)and proteolytic enzymes in the venom,and the activity of snake venom hyaluronidase was completely inhibited by the ethanol extract of Paris polyphylla ethanol.In addition,the ethanol extract of Paris polyphylla had a significant inhibitory effect on the coagulation enzyme activity of Agkistrodon venom.Conclusion The ethanol extract of Paris polyphylla extract inhibits effectively the activity of the main enzymes in Agkistrodon venom,which provides a theoretical basis for treating venomous snake bites.
