摘 要:目的:基于HPLC法及网络药理学预测龙胆(Gentiana scabra Bge.)质量标志物(quality markers,Q-marker),建立基于Q-marker的龙胆质量评价方法。方法:运用HPLC指纹图谱,筛选龙胆Q-marker的来源范围。通过网络药理学,确定其潜在Q-marker。体外实验验证其对氧化应激损伤的保护作用,确定龙胆的Q-marker。利用HPLC测定东北地区不同产地32批龙胆药材中Q-marker的含量,采用灰色关联度法(grey relation analysis,GRA)结合熵权法建立品质评价模型。结果:共标定了25个共有峰,指认出马钱苷酸、獐芽菜苦苷、龙胆苦苷和獐芽菜苷4个色谱峰。网络药理学显示4种化学成分均作用于TNF,EGFR,CASP3等21个靶点,通过丙型肝炎信号通路、TNF信号通路等发挥抗肝损伤、黄疸和目赤的药理作用。体外实验验证4个成分对氧化应激损伤的HepG2细胞均具有一定的保护作用,预测这4个成分是龙胆的Q-marker。32批样品中Q-marker含量的相对灰色关联系数在0.322~0.638,表明不同产地药材质量差异较大,且野生药材质量高于栽培药材。结论:本研究初步预测了龙胆的Q-marker,通过灰色关联度结合熵权法建立不同产地龙胆药材的质量评价模型,为完善该药材的质量评价体系提供参考。Objective:To predict the quality markers(Q-marker)of Gentiana scabra Bge.based on HPLC and network pharmacology,and establish a quality evaluation method for G.scabra Bge.Methods:HPLC fingerprint was used to screen the source range of G.scabra Bge.Q-markers.Network pharmacology was used to identify the potential Q-markers.In vitro experiments were performed to verify its protective effect on oxidative stress injury and determine the Q-markers of G.scabra Bge.HPLC was used to determine the contents of Q-markers in 32 batches of G.scabra Bge.from different producing areas in Northeast China.The grey correlation degree method combined with entropy weight method was used to establish the quality evaluation model.Results:A total of 25 common peaks were calibrated,and four chromatographic peaks of loganin,swertiamarin,gentiopicroside,and swertiamarin were identified.Network pharmacology showed that the four chemical components acted on 21 targets such as TNF,EGFR,CASP3,etc.,and played a pharmacological role in anti-liver injury,jaundice,and eye redness through hepatitis C signaling pathway and TNF signaling pathway.In vitro experiments verified that all the four components had a certain protective effect on HepG2 cells damaged by oxidative stress,and these four components were predicted to be Q-markers of G.scabra Bge.The relative grey correlation coefficients of Q-marker contents in 32 batches of samples ranged from 0.322 to 0.638,indicating that the quality of medicinal materials from different producing areas was greatly different,and the quality of wild medicinal materials was higher than that of cultivated medicinal materials.Conclusion:In this study,the Q-markers of G.scabra Bge.were preliminarily predicted,and the quality evaluation model of G.scabra Bge.from different regions was established by the grey correlation degree combined with entropy weight method,which provided a reference for improving the quality evaluation system of G.scabra Bge.
