摘 要:研究赛里木湖来源放线菌Streptomyces sp.A5的次级代谢产物。采用正相硅胶柱、Sephadex LH-20凝胶柱以及半制备高效液相色谱等分离方法,从该菌株的大米固体培养基发酵产物中分离得到5个化合物,通过NMR、MS等波谱方法鉴定为(±)-4′-methoxydehydroaltenusin(1)、alternariol 4-methyl ether(2)、6,8-dihydroxy-3-methylisocoumarin(3)、吲哚-3-甲酸甲酯(4)、streptcytosine H(5)。其中,化合物1为新的脱氢阿霉素类化合物,并对分离得到的化合物进行抗菌活性筛选。结果表明,化合物1对大肠杆菌、金黄色葡萄球菌以及白色念珠菌均呈现出中等的抑制活性,最小抑菌浓度(minimal inhibitory concentration,MIC)分别为12.5、6.25和6.25μg/mL,化合物2对这3种细菌表现出较弱抑制活性,其MIC均为25μg/mL。This study aims to investigate the secondary metabolites of Streptomyces sp.A5 derived from Sayram Lake.Five compounds were isolated from the solid fermentation extract of Streptomyces sp.A5 by using various chromatography methods including silica gel column,Sephadex LH-20 column and semipreparative HPLC.Their structures were determined by NMR,MS methods as a new dehydroaltenusin derivative,namely(±)-4′-methoxydehydroaltenusin(1),and four known compounds,including alternariol 4-methyl ether(2),6,8-dihydroxy-3-methylisocoumarin(3),indole-3-methyl formate(4),streptcytosine H(5).The antibacterial activities of compounds 1-5 were evaluated.The results indicated that compound 1 exhibited moderate antibacterial activity against Escherichia coli,Staphylococcus aureus and Candida albicans with minimal inhibitory concentration(MIC)at 12.5,6.25 and 6.25μg/mL,respectively.Compound 2 showed a weaker antibacterial activity against them with same MIC at 25μg/mL.
