摘 要:为研究温度对红鳍东方鲀(Takifugu rubripes)幼鱼生长和代谢的影响,以孵化12 d后的红鳍东方鲀幼鱼为研究对象,设置15℃(T15)、20℃(T20)和25℃(T25)三个温度梯度,养殖周期为30 d。试验结束后,测量不同处理组幼鱼的体长、体质量及存活率,并采集肝脏进行非靶向质谱联用法和液相色谱(LC—MS)代谢组学分析。结果表明,T25组体长和体质量以及特定生长率均显著高于T20组和T15组(P<0.05);T20组存活率高于T25组和T15组,T25处理组存活率高于T15组(P<0.05),其中T20组幼鱼存活率为84.81%,T25组为74.07%,T15组为64.44%。各温度对比组鉴定出的差异代谢产物数量分别为:T15-vs-T25有330种(117种上调、213种下调);T20-vs-T15有340种(208种上调、132种下调);T20-vs-T25有238种(77种上调、161种下调)。KEGG富集分析显示,差异代谢物主要参与氨基酸类生物合成与代谢、脂类代谢、蛋白质消化吸收、次级代谢产物生物合成以及三羧酸(TCA)循环等多种代谢通路。In order to investigate the influence of temperature on the growth and metabolism of Takifugu rubripes larvae,an experiment was conducted with larvae T.rubripes 12 days after hatching,three groups of larvae were reared for 30 d under 15℃(T15),20℃(T20),and 25℃(T25),respectively.At the end of the experiment,the body length,body weight,and survival rate of larvae in different treatment groups were measured,and the livers were collected for metabolomic analysis through LC—MS metabolomics analysis.The results showed that the body length and weight as well as the specific growth rate were significantly higher in the T25 group than in the T20 and T15 groups(P<0.05).The survival rate was higher in the T20 group than in the T25 and T15 groups,and it was higher in the T25 group than in the T15 group(P<0.05).The juvenile survival rate was 85%in the T20 group,74%in the T25 group and 64%in the T15 group.Metabolomics analysis of the liver revealed that 330 differential metabolites(117 up—regulated and 213 down—regulated)were identified in the T25 group compared to the T15 group.Compared with the T20 group,the 340 differential metabolites(208 up—regulated and 132 down—regulated)were identified in T15 group.Compared with the T20 group,340 differential metabolites(77 up—regulated,238 down—regulated)were identified in T25 group.The metabolite pathway enrichment analysis by KEGG showed that the differential metabolites were mainly involved in amino acid biosynthesis and metabolism,lipid metabolism,protein digestion and absorption,secondary metabolite biosynthesis,and the tricarboxylic acid(TcA)cycle pathways.
