摘 要:为了解高温、低氧以及高温低氧联合胁迫对刺参(Apostichopus japonicus)组织形态、抗氧化酶活性和HSP 70基因表达的影响,将刺参分别置于高温、低氧以及高温低氧共同胁迫的环境中进行试验,通过分析刺参胁迫后肠道组织切片,发现在高温、低氧以及共同胁迫下,肠道组织出现明显的退化;通过测定多种抗氧化酶活性发现,环境胁迫对T-AOC、GR、G-SH和SOD活性有明显影响(P<0.05),且在联合胁迫组变化尤为明显;HSP 70基因mRNA表达水平结果表明,HSP 70基因在高温、高温低氧联合胁迫下出现显著升高(P<0.05),在低氧胁迫下无显著变化(P>0.05)。综上所述,刺参对环境胁迫下会出现组织学、抗氧化酶和HSP 70 mRNA水平的响应,并在高温低氧联合胁迫下最为明显。To understand the impacts of high temperature,low dissolved oxygen and the combined stress on tissue morphology,antioxidant enzyme activity and HSP 70 gene expression in the sea cucumber Apostichopus japonicus,A.japonicus was exposed to high temperature,low dissolved oxygen,and the combined stress.By analyzing the intestinal tissue sections after stress,it was found that the intestinal tissue appeared obvious degradation under high temperature,low dissolved oxygen and the combined stress.It was also found that environmental stress had significant effects on the activities of T-AOC,GR,G-SH and SOD(P<0.05),especially in the combined stress group.The mRNA expression level of HSP 70 gene showed that HSP 70 gene increased significantly under high temperature and the combined stress(P<0.05),but there was no significant change under low dissolved oxygen(P>0.05).In conclusion,A.japonicus had a response to environmental stress in terms of histology,antioxidant enzymes and HSP 70 mRNA levels,and the changes were the most obvious under the combined stress of high temperature and low dissolved oxygen.
