摘 要:研究旨在为饲用芽孢杆菌的研发与应用提供参考。试验从肉鸡肠道内容物中分离筛选具有较强拮抗产气荚膜梭菌功能的芽孢杆菌,通过细菌形态学、生理生化鉴定和16S rDNA测序等方法确定其分类归属,并对其益生特性进行了研究。结果显示:菌株J5-8为地衣芽孢杆菌(Bacillus licheniformis),具有良好的耐热能力,经80、100、120℃处理10 min,芽孢存活率分别为94.6%、86.5%和81.1%。菌株J5-8在pH值低于3时仍有26%的菌体存活,在0.5%胆盐中存活率为82%,在模拟人工胃液、肠液等条件下均具有良好的耐受性。与同类菌株相比,菌株J5-8产酶种类及中性蛋白酶等活性均较高。研究表明,菌株J5-8具有良好的耐酸、胆盐以及耐温能力,而且能够产多种生物酶,可作为饲用芽孢杆菌进行防控产气荚膜梭菌的深入研究。The purpose of this study is to provide reference for the development and application of feeding bacillus.In this study,Bacillus with strong antagonistic function against Clostridium perfringens was isolated from the intestinal contents of broilers,and its classification was determined by morphological,physiological and biochemical identification and 16S rDNA sequencing,and its probiotic properties were studied.The results showed that strain J5-8 was Bacillus licheniformis with good heat resistance.After treatment at 80,100,and 120℃for 10 min,the spore-survival rate was 94.6%,86.5%,and 81.1%,respectively.Strains J5-8 still had 26%survival rate at pH below 3,and 82%survival rate in 0.5%bile salt.They were well tolerated under conditions such as simulated gastric juice and intestinal juice.Compared with the same strains,J5-8 had higher activity of producing enzymes and neutral protease.The results showed that strain J5-8 had good acid resistance,bile salt resistance and temperature resistance,and could produce a variety of biological enzymes,which could be used as feeding bacillus to further study the prevention and control of Clostridium perfringens.
