摘　　要：旨在观察猪正常和囊肿卵巢中血管生成素(ANG)的定位和表达情况,探讨ANG的表达与猪卵巢囊肿发生之间的关系。从屠宰场收集250~300日龄的大白猪母猪卵巢,根据形态将其分为正常组和卵巢囊肿组(n=8),通过HE染色对正常卵巢和囊肿卵巢进行形态学观察,利用Western blot检测卵巢中ANG蛋白的表达,采用免疫组织化学法检测卵巢中ANG的定位和表达情况。结果:外观上囊肿卵泡和囊肿黄体体积明显增大,卵泡膜细胞和黄体细胞均出现肿大、间隙不均和脂滴空泡增多的异常现象;Western blot结果表明囊肿卵巢中ANG的表达量极显著升高(P<0.01);免疫组化检测显示ANG在囊肿卵泡膜细胞和囊肿黄体细胞中的表达量明显上升,而在囊肿卵巢正常部位处有腔卵泡膜细胞中的表达量明显减少。结论:ANG与猪卵巢囊肿的发生之间存在关联,提示ANG可以作为一种诊断标志物,为临床上卵巢囊肿的确诊提供帮助。This study aimed to investigate the localization and expression of angiogenin(ANG) in normal and cystic ovaries of pigs,and to explore the relationship between ANG expression and the occurrence of ovarian cysts.Samples of ovaries from Large White sows aged 250-300 days were collected in a slaughterhouse,and they were divided into the normal and cystic groups(n=8) based on the morphological criteria.Then,histological observations of normal and cystic ovaries were conducted using HE staining.Finally,Western blot was used to detect ANG protein expression in the ovaries,and immunohistochemistry was employed to detect the localization and expression of ANG in the ovaries.The results showed that,morphologically,the cystic follicles and cystic corpora lutea were significantly enlarged,with abnormal phenomena such as hypertrophy of follicular theca cells and luteal cells,uneven gaps,and increased lipid droplets.The Western blot results showed that ANG expression in the cystic ovaries was significantly increased(P<0.01).The immunohistochemistry results indicated that ANG expression was markedly elevated in theca cells of the cystic follicles and the luteal cells of the cystic corpora lutea,while the expression of ANG in theca cells of the antral follicles in the normal parts of the cystic ovaries was also affected and significantly decreased.The present results indicated that there was association between ANG and the occurrence of ovarian cysts in pigs,suggesting that ANG could serve as a diagnostic marker to assist in the clinical diagnosis of ovarian cysts.
