摘 要:为研究鸭源海豚葡萄球菌(S.delphini)的生物学特征、药物敏感性及致病性,本研究从病鸭肝脏中通过分离纯化、革兰染色镜检、基质辅助激光解析电离飞行时间质谱(MALDI-TOF MS)鉴定、16S r RNA基因的PCR扩增与测序等方法进行细菌的分离鉴定;通过多重PCR鉴定分离菌株的亚群;分别通过k-B纸片法和雏鸭感染试验分析分离菌株的耐药性和致病性。结果显示,从患病鸭肝脏分离到产生β-溶血环的葡萄串状革兰阳性球菌;MALDI-TOF MS结果显示,分离菌株从蛋白水平上鉴定为S.delphini;16S r RNA基因的PCR扩增及序列进化树分析结果显示,扩增出1135 bp的目的条带,与S.delphini聚为一个进化分支,表明分离菌株从基因水平上鉴定为S.delphini;多重PCR鉴定结果显示,该菌为S.delphini A亚群;药敏试验结果显示,分离菌株对四环素类药物的多西环素、头孢菌素类药物的头孢吡肟、β-内酰胺类药物的青霉素G等敏感;对氨基糖苷类药物的大观霉素和庆大霉素、头孢菌素类药物的头孢唑林和头孢他啶等中介;对喹诺酮类药物的氧氟沙星、大环内酯类药物的红霉素、氯霉素类药物氟苯尼考等耐药,是典型的多重耐药菌。雏鸭感染试验结果显示,分离菌对雏鸭有一定的致死性,能引起其肝脏不同程度颜色变浅、出血及坏死点,腹腔含少量混浊、黄色浆液,膀胱有轻微化脓;本研究首次从鸭体内分离到S.delphini A亚群,该菌株对雏鸭有一定的致病性,且呈多重耐药,因此其带来的潜在风险不可忽视,本研究结果为该菌进一步的研究和防控其的感染提供参考。To investigate the biological characteristics,drug sensitivity and pathogenicity of Staphylococcus delphini from duck,the bacteria were isolated and identified from the liver of diseased ducks by isolation and purification,Gram staining microscopy,matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS),and 16S rRNA gene sequence analysis.The subtypes of the isolated strain were identified by multiplex PCR.The drug resistance and pathogenicity of the isolate were detected by drug sensitivity test and duckling infection test,respectively.The results showed that grape striated positive cocci producingβ-hemolytic ring were isolated from the liver of diseased ducks.MALDI-TOF MS results showed that the isolated strain was identified as Staphylococcus delphini at the protein level.PCR amplification of 16S rRNA gene and phylogenetic tree analysis showed that the isolated strain was identified as Staphylococcus delphini at the genetic level.Multiplex PCR identification showed that the bacteria were classified as Staphylococcus delphini subgroup A.The infection test of ducklings showed that the isolate was lethal to ducklings to a certain extent,and could cause different degrees of pale color of liver,bleeding and necrotic points,and the abdominal cavity contained a small amount of increased turbidity,yellow and serous fluid,and the bladder was slightly suppurative.The results of drug sensitivity test showed that the isolate was sensitive to doxycycline of tetracyclines,cefepime of cephalosporins,and penicillin G ofβ-lactams.It is moderately sensitive to daguanomycin and gentamicin of aminoglycosides,cefazolin and ceftazidime of cephalosporins.It is a typical multi-drug resistant strain,which is resistant to ofloxacin of quinolones,erythromycin of macrolides,and florfenicol of chloramphenicol.In this study,a stain of Staphylococcus delphini subtype A was isolated from ducks for the first time,the isolate demonstrates pathogenicity towards ducklings and exhibits multiple drug resistance,
