摘  要:目的基于网络药理学探索痰热清注射液致不良反应的分子机制。方法通过中药系统药理学数据库分析平台(TCMSP)收集痰热清注射液的化合物,在PharmMapper数据库中预测可能的靶点,与GeneCards数据库中不良反应相关靶点相映射,获得痰热清注射液导致不良反应发生的可能作用靶点;对这些靶点进行PPI网络图的构建、GO富集分析和通路分析以揭示痰热清注射液导致不良反应发生的可能作用机制。结果通过TCMSP共筛选出44个化合物,449个靶点,其中致不良反应相关靶点26个;PPI网络图显示ALB、ACE、HMOX1、PPARG是4个联系最多的靶点,涉及对外源性刺激的反应、外源分解代谢过程、ERK1和ERK2级联的调节等生物过程,作用机制与调控癌症通路、药物代谢细胞色素P450、代谢途径等生物学通路有关。结论痰热清注射液致不良反应的主要成分有芹菜素7 O葡萄糖苷、白杨素、芹菜素等,ALB、ACE、HMOX1、PPARG是其主要作用靶点。Objective To explore the molecular mechanism of adverse reactions caused by Tanreqing injection based on network pharmacology.Methods The compounds of Tanreqing injection were collected from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the potential targets were predicted in PharmMapper database and mapped with the related targets of adverse reactions in GeneCards database to obtain potential targets of Tanreqing injection causing adverse reactions.PPI network diagrams were constructed,and GO enrichment analysis and pathway analysis were performed on these targets to reveal the possible mechanism of adverse reactions caused by Tanreqing injection.Results A total of 44 compounds and 449 targets were screened by TCMSP,including 26 targets related to adverse reactions.The PPI network diagram showed that ALB,ACE,HMOX1,and PPARG were the four most-connected targets,involving biological processes such as response to exogenous stimuli,exogenous catabolic processes,and regulation of ERK1 and ERK2 cascade.The mechanism of action was related to the regulation of pathways in cancer,drug metabolism-cytochrome P450,metabolic pathways,and other biological pathways.Conclusion The main components of adverse reactions caused by Tanreqing injection include apigenin-7-O-glucoside,chrysin,apigenin,etc.ALB,ACE,HMOX1,and PPARG are its main targets of action.
