摘 要:目的基于中医“异病同治”理论,探讨温脾汤(WPD)治疗慢性肾衰竭(CRF)、肠易激综合征(IBS)、消化性溃疡(PU)的作用机制。方法运用TCMSP数据平台获取WPD的化学成分及靶点,检索GeneCards等数据库提取CRF、IBS和PU的靶点,利用Venny软件得出药物、疾病交集靶点。采用Cytoscape 3.7.2软件制作WPD治疗CRF、IBS和PU的药物成分靶点疾病网络分布图,以STRING平台构建WPD共同靶点的蛋白质蛋白质相互作用(PPI)网络,基于BioGPS数据平台和Cytoscape 3.7.2软件构建“WPD关键靶点器官组织”网络分布图,借助DAVID数据库进行GO功能和KEGG通路富集分析。结果预测出WPD共有70个交集靶点发挥异病同治CRF、IBS和PU的作用。PPI网络筛选出TNF、TP53等可能是WPD治疗CRF、IBS和PU的关键靶点。GO功能共富集323个条目,主要涉及基因表达正调控等因素;KEGG共富集119条通路,包括TNF、HIF-1等信号通路。芦荟大黄素、山柰酚等活性成分与TP53、TNF等核心靶点分子对接结果良好。结论WPD通过多组分、多靶点和多途径的特点治疗CRF、IBS和PU,为中医“异病同治”理论提供新的思路,同时也为WPD的临床应用及机制研究提供了生物信息学依据。Objective To explore the mechanism of action of Wenpi decoction(WPD)in treating chronic renal failure(CRF),irritable bowel syndrome(IBS)and peptic ulcer(PU)based on the traditional Chinese medicine theory of“treating different diseases with the same method”.Methods The TCMSP data platform was employed to identify the active ingredients and targets of WPD,GeneCards and other databases were utilized to acquire the targets of CRF,IBS and PU,and Venny software was used to obtain the intersection targets of drugs and diseases.Cytoscape 3.7.2 software was used to map the drug component target disease network diagram of WPD for treating CRF,IBS,and PU.STRING platform was used to construct protein-protein interaction(PPI)networks of the common targets of WPD.BioGPS data platform and Cytoscape 3.7.2 software were employed to construct the network diagram of“key targets of WPD organ tissue”.DAVID database was utilized to perform GO functional enrichment analysis and KEGG pathway enrichment analysis.Results A total of 70 intersection targets of WPD were predicted to play the role of treating different diseases with the same method for CRF,IBS and PU.PPI network screened TNF and TP53 as the key targets of WPD for the treatment of CRF,IBS and PU.A total of 323 items were enriched in GO function,mainly involving positive regulation of gene expression and other factors.A total of 119 pathways were enriched in KEGG,including TNF,HIF-1 and other signaling pathways.Active ingredients such as aloe-emodin and kaempferol were in good molecular docking with core targets such as TP53 and TNF.Conclusion WPD treats CRF,IBS and PU through the characteristics of multi-component,multi-target,and multi-pathway,which provides new ideas for the theory of“treating different diseases with the same method”in traditional Chinese medicine,and also provides a bioinformatics basis for the clinical application and mechanism research of WPD.
