摘  要:目的研究芪参益气滴丸对心肌缺血再灌注损伤的保护作用及作用机制。方法将心肌细胞随机分为5组,其中A组为正常H9c2细胞+空白血清组,B组为OGD/R+空白血清组,C组为OGD/R+低剂量含药血清组,D组为OGD/R+中剂量含药血清组,E组为OGD/R+高剂量含药血清组,按照分组给予相应的药物干预,LDH检测细胞培养液毒性;CCK-8法检测细胞活性;流式细胞仪检测细胞凋亡;ELISA检测炎症因子浓度;WB检测p-MST、p-YAP、p-TAZ、TEAD1的表达量。结果与B组心肌细胞比较,C组、D组和E组的心肌细胞活性增加,凋亡数量减少,培养液细胞毒性降低,炎症因子含量降低,p-MST、p-YAP、p-TAZ蛋白表达降低,而TEAD1蛋白表达升高(P<0.01)。结论芪参益气滴丸对糖氧剥夺/复氧H9c2心肌细胞的保护作用与Hippo通道关系密切。Objective:To study the protective effect and mechanism of action of Qishen Yiqi Dropping Pills on myocardial ischaemia-reperfusion injury.Methods:Cardiomyocytes were randomly divided into 5 groups,in which group A was normal H9c2 cells+blank serum group,group B was OGD/R+blank serum group,group C was OGD/R+low-dose drug-containing serum group,group D was OGD/R+medium-dose drug-containing serum group,and group E was OGD/R+high-dose drug-containing serum group.Corresponding drug interventions were given according to the grouping,and the toxicity of the cell culture fluid was detected by LDH;CCK-8 assay was used to detect cell activity;flow cytometry was used to detect apoptosis;ELISA was used to detect the concentration of inflammatory factors;WB was used to detect the expression of p-MST,p-YAP,p-TAZ,TEADI.Results:Compared with cardiomyocytes in group B,cardiomyocytes in groups C,D and E showed increased activity,decreased apoptosis,decreased cytotoxicity of the culture medium,decreased inflammatory factor content,decreased expression of p-MST,p-YAP,p-TAZ proteins,and elevated expression of TEAD1 proteins,and the difference was statistically significant(P<0.01).Conclusion:The protective effect of Qishen Yiqi Dropping Pills on glyoxylate deprivation/reoxygenation H9c2 cardiomyocytes is closely related to Hippo channel.
