摘  要:以腺苷、生物节律肽和酵母发酵液制备了一种功效组合物,探讨了该组合物对皮肤昼夜节律和屏障功能的影响。采用蓝光对人永生化角质细胞(HaCaT)进行光处理以模拟表皮熬夜环境,构建细胞测试模型。检测了处理前后昼夜节律蛋白,屏障相关蛋白的表达水平,结果显示体积分数1%的功效组合物能显著提高Ha Ca T中脑和肌肉组织芳香烃受体类似蛋白1(P<0.05)、节律运动输出周期蛋白(P<0.0001)、水通道蛋白3(P<0.01)、聚丝蛋白(P<0.01)和兜甲蛋白(P<0.001)的表达。人体皮肤测试表明:连续使用含有该功效组合物的护肤品28天后,皮肤经皮水分散失降低了11.29%(P<0.01),角质层含水量提高了35.60%(P<0.001)。上述结果说明该组合物能调节皮肤昼夜节律并增强皮肤屏障功能。A functional composition was prepared with adenosine,Chronocyclin^(R)and Revitalin^(R)PF.The effects of the functional composition on circadian rhythm and skin barrier function were investigated.A cell model was constructed by treating HaCaT cells with blue light to simulate sleep deprivation.The expression of circadian rhythm proteins and barrier related proteins were detected before and after treatment.The results showed that 0.1%(in volume fraction)functional composition significantly increased the expression of brain and muscle arnt-like protein-1(P<0.05),circadian locomotor output cycles kaput protein(P<0.0001),aquaporin 3(P<0.01),filaggrin(P<0.01)and loricrin(P<0.001)in HaCaT.In addition,human skin tests showed that transepidermal water loss was reduced by 11.29%(P<0.01)and stratum corneum hydration was in creased by 35.60%(P<0.001)after 28 d continuous use of the cosmetics containing the functional composition.These results indicate that the functional composition could regulate the circadian rhythm and enhance the skin barrier.
