摘 要:目的:探讨麝香通心滴丸(Shexiang Tongxin Dropping Pills,STDP)通过靶向Keap1-Nrf2/ARE(Kelch样ECH相关蛋白1-核因子E2相关因子2/抗氧化反应元件信号通路)信号通路对H9C2心肌细胞H_(2)O_(2)过氧化损伤的保护作用及其机制。方法:采用H_(2)O_(2)诱导H9C2心肌细胞氧化应激损伤模型,实验分为正常对照组(Control)、H_(2)O_(2)组(H_(2)O_(2))、麝香通心滴丸组(STDP),H_(2)O_(2)组构建H9C2细胞过氧化损伤模型,麝香通心滴丸组在构建H_(2)O_(2)过氧化损伤前给予50 mg/L麝香通心滴丸预处理12 h。CCK8法检测各组细胞活力值,DCFH-DA荧光探针法检测各组细胞内ROS水平,Western blot检测各组H9C2心肌细胞Keap1-Nrf2/ARE通路相关蛋白表达。结果:与正常对照组比较,H_(2)O_(2)组细胞活力值降低(P<0.05),细胞内ROS水平升高,Nrf2、HO-1、NQO1蛋白表达下调,Keap1蛋白表达上调,差异均有统计学意义(均P<0.05)。与H_(2)O_(2)组比较,麝香通心滴丸组细胞活力值升高(P<0.05),ROS水平降低,Nrf2、HO-1、NQO1蛋白表达上调,Keap1蛋白表达下调,差异均有统计学意义(均P<0.05)。结论:麝香通心滴丸可以通过靶向Keap1-Nrf2/ARE信号通路增强H9C2心肌细胞的抗氧化能力,发挥对H_(2)O_(2)心肌细胞过氧化损伤的保护作用。Objective:To investigate the protective effects and mechanisms of Shexiang Tongxin Dropping Pills(STDP)on H_(2)O_(2)-induced oxidative damage in H9C2 cardiomyocytes by targeting the Keap1-Nrf2/ARE signaling pathway.Methods:An oxidative stress model was established in H9C2 cardiomyocytes induced by H_(2)O_(2).The cells were divided into the control group(Control),H_(2)O_(2)group(H_(2)O_(2)),and STDP group(STDP).The H_(2)O_(2)group was used to construct the oxidative damage model in H9C2 cells,and the STDP group was pretreated with 50 mg/L STDP for 12 hours before constructing the H_(2)O_(2)oxidative damage model.Cell viability was detected by CCK8 assay,intracellular ROS levels were measured by the DCFH-DA fluorescent probe method,and protein expressions related to the Keap1-Nrf2/ARE pathway were detected by Western blot.Results:Compared with the control group,the cell viability in the H_(2)O_(2)group decreased(P<0.05),intracellular ROS levels increased,expressions of Nrf2,HO-1,and NQO1 proteins decreased,and Keap1 protein expression increased,all with statistically significant differences(P<0.05).Compared with the H_(2)O_(2)group,the STDP group showed increased cell viability(P<0.05),decreased ROS levels,increased expressions of Nrf2,HO-1,and NQO1 proteins,and decreased Keap1 protein expression,all with statistically significant differences(P<0.05).Conclusion:Shexiang Tongxin Dropping Pills can protect H9C2 cardiomyocytes from H_(2)O_(2)oxidative damage by targeting the Keap1-Nrf2/ARE signaling pathway,thereby enhancing the antioxidant capacity of H9C2 cardiomyocytes.
