益气养阴消癥方对糖尿病肾病大鼠肾功能及相关炎症因子影响的实验研究

摘　　要：目的:探讨益气养阴消癥方对糖尿病肾病(diabetic kidney disease,DKD)大鼠肾功能及相关炎症因子的影响。方法:SD大鼠60只,随机分为正常对照组、模型对照组、阳性对照组(缬沙坦8 mg/kg)和益气养阴消癥方高(8 g/kg)、中(4 g/kg)、低(2 g/kg)剂量组,每组10只大鼠。除正常组外,其他各组大鼠采用链脲佐菌素腹腔注射(50 mg/kg)建模,成模后各组大鼠给予相应药物灌胃,连续给药8周。观察各组大鼠一般状况,检测各组大鼠24 h尿蛋白定量及血尿素氮(BUN)、肌酐(SCr)水平,ELISA检测血清IL-1β(白介素-1β)和IL-6(白介素-6)含量。结果:与正常组比较,模型组大鼠一般状况差,24 h尿蛋白定量和血BUN、SCr及血清IL-1β、IL-6水平明显升高(均P<0.05或P<0.01);各给药组一般状况较模型组明显改善,24 h尿蛋白定量和血BUN、SCr及血清IL-1β、IL-6水平明显降低(均P<0.05);益气养阴消癥方高剂量组效果与阳性对照组相当(均P>0.05)。结论:益气养阴消癥方可保护DKD大鼠肾功能,抑制炎症因子分泌。Objective:To explore the effects of Yiqi Yangyin Xiaozheng Decoction on renal function and inflammatory cytokines of rats with diabetic kidney disease.Methods:60 rats were divided into normal control group,model group,positive group(Valsartan,8 mg/kg)and Yiqi Yangyin Xiaozheng Decoction high dose(8 g/kg),middle dose(4 g/kg),low dose(2 g/kg)group,each with 10 rats.Except normal group,rats in the other groups were modeled by STZ(50 mg/kg)intraperitoneal injection,then each group was given the corresponding medicine by Ig for 8 weeks,normal and model groups rats were given the same volume distilled water.The general condition of each group rats were observed,24 h urinary protein,blood BUN and SCr levels were examined,ELISA was used to detect serum IL-1βand IL-6 levels.Results:Compare with normal group,the general condition of model group rats were bad,24 h urinary protein,blood BUN and SCr,serum IL-1βand IL-6 levels were raised significantly(P<0.05 or P<0.01);the general condition of each medication group rate were improve obviously,24 h urinary protein,blood BUN and SCr,serum IL-1βand IL-6 levels were lowered significantly(P<0.05),the effects of Yiqi Yangyin Xiaozheng Decoction high dose group were similar to those of positive group(P>0.05).Conclusion:Yiqi Yangyin Xiaozheng Decoction can protect renal function of rats with diabetic kidney disease,inhibit inflammatory.
