摘  要:目的:观察胆木提取物对哮喘大鼠气道高反应性和炎症的影响。方法:卵白蛋白反复攻击7 d制备大鼠哮喘模型,实验分正常组、模型组、胆木提取物(100、300、900 mg/kg)组和地塞米松(DXM 2 mg/kg)组,每组8只,各组分别给予相应剂量药物连续灌胃给药10 d。用乙酰甲胆碱(Mch)激发哮喘大鼠的气道高反应性,检测各组大鼠的肺阻力(RL)和动态肺顺应性(C_(dyn));取支气管肺泡灌洗液(BALF)检测气道炎症细胞的浸润情况;同时取大鼠肺组织行HE染色,显微镜下观察肺组织嗜酸性粒细胞浸润情况。结果:与正常组比较,模型组大鼠气道反应性明显增高,同时BALF中白细胞总数、嗜酸性粒细胞数目显著增加,肺组织嗜酸性粒细胞明显浸润至支气管周围和血管周围。胆木提取物各组呈剂量依赖地抑制大鼠的气道高反应性,降低BALF中白细胞总数、嗜酸性粒细胞数目,减轻肺组织嗜酸性粒细胞浸润。结论:胆木提取物对大鼠支气管哮喘有防治作用,为其应用于哮喘的治疗提供了实验依据。Objective:To observe the effects of Nauclea Officinalis Extract on airway hyperresponsiveness and airway inflammation responses in asthmatic rats.Methods:The asthma model was made by repeatedly attacking with ovalbumin for 7 days on ovalbumin sensitized rats.The experimental rats were divided into normal group,model group,Nauclea Officinalis Extract group(100,300,900 mg/kg)and Dexamethasone group(DXM 2 mg/kg),each group contained 8 rats.Each group rats were subjected to continuous intragastric administration for 10 days based on the corresponding dose,then were stimulated airway hyperresponsiveness(AHR)with methacholine(Mch)and measured lung resistance(RL)and dynamic lung compliance(C_(dyn));the infiltration of inflammatory cells in the airways were detected by bronchoalveolar lavage fluid(BALF);HE staining was performed on lung tissues of rats,and the infiltration of pulmonary eosinophilics were observed under microscope.Results:Compared with the normal group,the airway reactivity of the model group was significantly increased,while the total number of white blood cells and eosinophils in BALF were significantly increased,and eosinophils in lung tissue infiltrated into the bronchus and blood vessels.Nauclea Officinalis Extract inhibited airway hyperreactivity,lowered the total number of white blood cells and eosinophils in BALF,reduced the infiltration of eosinophils in lung tissue,The effects showed dose dependence.Conclusion:Nauclea Officinalis Extract can prevent and cure bronchial asthma in rats,which provides experimental basis for its application in the treatment of asthma.
