摘 要:目的:探究参芪地黄汤对长双歧杆菌底物利用能力及代谢物的影响。方法:将长双歧杆菌分为空白组、对照组、给药组;通过生长曲线观察参芪地黄汤最大无毒浓度;采用Biology-ECO技术探究参芪地黄汤水煎液对长双歧杆菌利用碳源底物能力的影响,并基于液质联用技术分析差异代谢物。结果:参芪地黄汤水煎液最大给药量为0.84 g/mL;与对照组相比,给药组对糖类及其衍生物、氨基酸类、多聚物类以及胺类的利用能力均无显著性差异(P>0.05);对羧酸类、α-环式糊精以及2-羟基苯甲酸的利用能力均具有显著性差异(P<0.05)。共检测出差异代谢物24种,其中16种给药后上调,8种下调。结论:参芪地黄汤对长双歧杆菌底物利用及代谢物均有一定的影响。Objective:To investigate the effects of Shenqi Dihuang Decoction on substrate utilization capacity and metabolism of Bifidobacterium longum.Methods:Bifidobacterium longum was divided into blank group,control group and drug administration group.The maximum non-toxic concentration of Shenqi Dihuang Decoction was observed by the growth curve.Biology-ECO technique was used to investigate the effect of Shenqi Dihuang Decoction on the ability of Bifidobacterium longum to utilize carbon source substrate,and the differential metabolites were analyzed based on UPLC-Q-TOF-MS technique.Results:The maximum dosage of Shenqi Dihuang Decoction was 0.84 g/mL;Compared with control group,there were no significant differences in the utilization capacity of carbohydrates and their derivatives,amino acids,polymers and amines in drug administration group(P>0.05).The utilization capacity of p-carboxylic acids,α-cyclodextrin and 2-hydroxybenzoic acid was significantly different(P<0.05).A total of 24 kinds of different metabolites were detected,of which 16 were up-regulated and 8 were down-regulated after administration.Conclusion:Shenqi Dihuang Decoction has effects on substrate utilization and metabolism of Bifidobacterium longum.
