百香果茎基腐病菌拮抗菌的筛选、鉴定及其生防效果

摘  要:以百香果茎基腐病菌优势菌腐皮镰孢菌Fusarium solani为靶标,从百香果健康根、茎、叶和花中分离纯化内生细菌,开展对峙培养,及离体叶片、盆栽种苗接种防效试验,筛选鉴定防治效果明显的拮抗菌株,对菌株进行形态学分析,采用16S rRNA和gyrB基因序列比对,及其序列构建的系统发育进化树进行分子鉴定,并测定其拮抗菌胞外酶活性。结果表明,菌株EB-G-11-01对百香果茎基腐病菌的菌丝生长具有明显的抑制效果,抑制率为53.2%;离体叶片和种苗盆栽试验中,菌株EB-G-11-01对百香果茎基腐病平均防效分别达到44.23%和95.39%,均高于枯草芽孢杆菌1000倍液的平均防效26.17%和78.45%。鉴定菌株EB-G-11-01为枯草芽孢杆菌Bacillus subtilis,其能产生胞外蛋白酶、β-1,3-葡聚糖酶和纤维素酶,具有作为生防菌开发利用的潜力。Fusarium solani,the dominant specie of stem rot disease pathogen of passion fruits was used as the target,and the endogenous bacteria were isolated and purified from the healthy roots,stems,leaves and flowers of passion fruits for confrontation culture and inoculation test of in vitro leaves and potted seedlings,also for screening and identification antagonistic bacterial strains.The morphology of isolated strains was analyzed.The molecular identification of the strain was conducted through 16S rRNA and gyrB gene sequence alignment,and phylogenetic tree construction based on the sequences.The extracellular enzyme activity of the antagonistic bacteria was determined.The results showed that the inhibitory rate of EB-G-11-01 against F.solani was 53.2%.The average control efficiency of EB-G-11-01 by in vitro inoculation and pot experiment was 44.23%and 95.39%,respectively,both higher than that of Bacillus subtilis WB1000,which was 26.17%and 78.45%,respectively.The strain EB-G-11-01 was identified as B.subtilis.It could produce extracellular protease,β-1,3-glucanase and cellulase,which has the potential to be used as biocontrol bacteria.
