摘 要:以“台农1号”紫果百香果嫩茎为外植体,改良RMS添加7 g/L琼脂、25 g/L蔗糖和0.5 g/L花宝1号为基本培养基,添加不同植物生长调节剂直接诱导腋芽萌发成苗培养,再采用热处理结合茎尖培养进行脱毒处理,得到的组培苗检测黄瓜花叶病毒(CMV)、西番莲木质化病毒(PWV)、西番莲斑驳病毒(PaMV)、西番莲扭叶病毒(PLDV)和东亚西番莲病毒(EAPV),选取病毒检测阴性组培苗进行芽增殖及生根培养,优化不同培养阶段植物生长调节剂种类、浓度和配比。结果表明,RMS+2.5 mg/L玉米素适合丛生芽诱导,诱导率达到96%;RMS+1.5 mg/L玉米素+0.2 mg/Lα-萘乙酸+0.1 mg/L GA3适合继代增殖培养,增殖系数达3.43;RMS+1.0 mg/L吲哚丁酸+0.2 mg/Lα-萘乙酸适合生根诱导,生根率达93%。热处理结合茎尖培养得到的组培苗经过病毒复检后发现,20份样品都未检测出CMV、PWV、PaMV、PLDV和EAPV,5种病毒的脱毒率达100%,突破了百香果脱毒种苗的快速繁育技术瓶颈。With young stems of purple-fruited passion fruit Tainong No.1 as explants,the modified RMS with addition of 7 g/L agar,25 g/L sucrose and 0.5 g/L Huabao 1 as basic medium,the germination of axillary buds was directly induced by adding different plant hormones.The virus-free plantlets were generated through virus elimination process by heat treatment combined with stem tip culture.The seedlings were detected for passion fruit cucumber Mosaic virus(CMV),passion fruit lignification virus(PWV),passion fruit mottling virus(PaMV),passion fruit leaf twist virus(PLDV)and East Asian passion fruit virus(EAPV).The virus negative buds were selected for multiplication and rooting culture,and optimizing the growth regulators,concentrations and compositions in different culture stages.The results showed that RMS+2.5 mg/L ZEA was suitable for inducing clustered buds,with an induction rate of 96%.RMS+1.5 mg/L ZEA+0.2 mg/L NAA+0.1mg/L GA 3 was suitable for proliferation subculture,with a proliferation coefficient of 3.43.RMS+1.0 mg/L IBA+0.2 mg/L NAA was suitable for root induction,with a rooting rate of 93%.CMV,PWV,PaMV,PLDV and EAPV were not detected in 20 samples of tissue cultured seedlings obtained through heat treatment combined with stem tip culture,with virus elimination rate of these 5 viruses reaching 100%,archived the breakthrough for the bottleneck of rapid propagation technology for virus-free passion fruit seedlings.
