摘  要:采用硅胶柱色谱、HP20柱色谱、聚酰胺柱色谱、反相C18柱色谱、制备型高效液相色谱等多种色谱分离方法对刺黄花根的化学成分进行系统分离和纯化,并通过理化性质和波谱学手段(1D NMR、2D NMR、UV、MS、CD)鉴定化合物结构。从刺黄花根甲醇提取物中分离得到4个苯丙素类化合物,分别鉴定为(2R)-1-(4-羟基-3,5-二甲氧基苯基)-1-丙酮-O-β-D-葡萄糖苷(1)、丁香酸甲酯(2)、(+)-丁香脂素(3)、丁香脂素-4-O-β-D-吡喃葡萄糖苷(4)。其中化合物1为新化合物,其余化合物均为首次从该植物中分离得到。通过测定脂多糖(LPS)诱导的小鼠RAW264.7巨噬细胞培养液中NO的释放量,评价化合物对炎症反应的抑制作用。在给药浓度为10μmol·L^(-1)时,4个化合物均可抑制LPS诱导的RAW264.7细胞中NO的释放,发挥抗炎作用。The chemical constituents were systematically separated from the roots of Berberis polyantha by various chromatographic methods,including silica gel column chromatography,HP20 column chromatography,polyamide column chromatography,reversed-phase C_(18)column chromatography,and preparative high-performance liquid chromatography.The structures of the compounds were identified by physicochemical properties and spectroscopic techniques(1D NMR,2D NMR,UV,MS,and CD).Four phenylpropanoids were isolated from the methanol extract of the roots of B.polyantha,and they were identified as(2R)-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone-O-β-D-glucopyranoside(1),methyl 4-hydroxy-3,5-dimethoxybenzoate(2),(+)-syringaresinol(3),and syringaresinol-4-O-β-D-glucopyranoside(4).Compound 1 was a new compound,and other compounds were isolated from this plant for the first time.The anti-inflammatory activity of these compounds was evaluated based on the release of nitric oxide(NO)in the culture of lipopolysaccharide(LPS)-induced RAW264.7 macrophages.At a concentration of 10μmol·L^(-1),all the four compounds inhibited the LPS-induced release of NO in RAW264.7 cells,demonstrating potential anti-inflammatory properties.
