摘  要:目的:探讨汉黄芩素对乙型肝炎病毒(HBV)大鼠肝功能的影响及其作用机制。方法:斯泼累格·多雷(Sprague-Dawley,SD)雄性大鼠72只,随机分为对照组,HBV模型组,IFN-α组,汉黄芩素低、中、高剂量组,每组12只。除对照组,其余大鼠采用尾静脉注射HBV抗原构建HBV大鼠模型。IFN-α组大鼠高压尾静脉注射10μg pKCMvint.IFN-α,汉黄芩素低、中、高剂量组大鼠分别腹腔注射汉黄芩素10 mg/kg、20 mg/kg、40 mg/kg,对照组和模型组大鼠腹腔注射生理盐水10 mL/(kg·d),持续2周。2周后处死,酶联免疫吸附试验(ELISA)法测定大鼠血清HBV标志物乙型肝炎e抗原(HBeAg)、乙型肝炎表面抗原(HBsAg)和肝功能指标谷丙转氨酶(GPT)、谷草转氨酶(GOT)水平,苏木精-伊红(HE)染色和马松(Masson)染色分别检测肝组织病理学改变和纤维化情况,蛋白免疫印迹(Western blotting)检测肝组织匀浆中磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)/哺乳动物雷帕霉素靶蛋白(mTOR)通路蛋白表达。结果:与HBV模型组比较,汉黄芩素中、高剂量组大鼠饮食和排泄基本正常,腹腔积液消失,肝组织损伤缓解,肝纤维化减少,血清HBeAg、HBsAg、GOT、GPT水平和肝组织p-PI3K/PI3K、p-AKT/AKT、p-mTOR/mTOR蛋白水平显著降低(均P<0.05)。结论:汉黄芩素可保护HBV大鼠肝功能,其可能的机制是抑制PI3K/AKT/mTOR信号通路。Objective:To investigate the effects and underlying mechanisms of wogonin on liver function in hepatitis B virus(HBV)model rats.Methods:Seventy-two male Sprague-Dawley(SD)rats were randomly divided into six groups(n=12 per group):control,HBV model,IFN-α,and low-,medium-,and high-dose wogonin groups.Except for the control group,all rats were injected with HBV antigen via the tail vein to establish the HBV model.The IFN-αgroup received a high-pressure tail vein injection of 10μg of pKCMvint.IFN-α,while the wogonin groups received intraperitoneal injections of 10 mg/kg,20 mg/kg,or 40 mg/kg wogonin,respectively.The control and model groups received normal saline(10 mL/kg·d)intraperitoneally for two weeks.After two weeks,the rats were sacrificed,and serum HBV markers,including hepatitis B e-antigen(HBeAg)and hepatitis B surface antigen(HBsAg),as well as liver function indicators alanine aminotransferase(GPT)and aspartate aminotransferase(GOT),were measured using enzyme-linked immunosorbent assay(ELISA).Hematoxylin-eosin(HE)and Masson staining were used to assess liver histopathological changes and fibrosis.Western blot was performed to analyze the expression of phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/mammalian target of rapamycin(mTOR)signaling pathway proteins in liver tissue homogenates.Results:Compared with the HBV model group,the medium-and high-dose wogonin groups showed improved dietary intake and excretion,disappearance of ascites,alleviation of liver tissue damage,and reduced liver fibrosis.Serum levels of HBeAg,HBsAg,GOT,and GPT,as well as liver tissue p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR protein levels,were significantly decreased(P<0.05).Conclusion:Wogonin protects liver function in HBV model rats,potentially through inhibition of the PI3K/AKT/mTOR signaling pathway.
