摘  要:目的 探究血必净注射液对脓毒症诱导的小鼠急性肺损伤的改善作用及机制。方法 C57BL/6小鼠随机分为对照组(Control组)、脓毒症急性肺损伤组(ALI组)和血必净组(XBJ组),每组10只,采用盲肠结扎穿孔术建立脓毒症急性肺损伤模型。观察各组小鼠精神状态、活动、饮食、呼吸及对刺激的反应;取各组小鼠肺组织称量湿重和干重并计算肺湿重/干重比;HE染色观察各组小鼠肺组织病理损伤;ELISA检测各组小鼠肺组织中IL-1β和IL-18含量;Western blot检测各组小鼠肺组织中NLRP3、pro-caspase-1、cleaved caspase-1、GSDMD-N、p-c-Jun和c-Jun蛋白的表达情况。结果 血必净降低脓毒症急性肺损伤小鼠肺湿重/干重比,改善小鼠肺组织病理损伤,降低小鼠肺组织中IL-1β和IL-18含量,下调小鼠肺组织中NLRP3、ASC、pro-caspase-1、cleaved caspase-1、GSDMD-N、p-c-Jun和c-Jun蛋白表达。结论 血必净改善脓毒症诱导的急性肺损伤和肺组织的炎症反应,其机制与下调c-Jun表达并抑制其激活,从而抑制NLRP3炎症小体激活和细胞焦亡有关。Objective To explore the improvement effect and mechanism of Xuebijing injection on acute lung injury induced by sepsis in mice.Methods C57BL/6 mice were randomly divided into control group(Control group),septic acute lung injury group(ALI group)and Xuebijing group(XBJ group),with 10 mice in each group.Acute lung injury model of sepsis was established by cecal ligation and perforation.Observe the mental state,activity,diet,breathing and response to stimulation of mice in each group.The lung tissues of mice in each group were weighed for wet weight and dry weight,and the ratio of wet weight to dry weight was calculated.HE staining was used to observe the pathological injury in lung tissue of mice in each group.ELISA was used to detect the contents of IL⁃1βand IL⁃18 in lung tissue of mice in each group.Western blot was used to detect the expressions of NLRP3,pro⁃caspase⁃1,cleaved caspase⁃1,GSDMD⁃N,p⁃c⁃Jun and c⁃Jun proteins in lung tissues of mice in each group.Results Xuebijing can reduce the wet/dry weight ratio of lung in mice with acute lung injury due to sepsis,improve the pathological injury of lung tissue,reduce the contents of IL⁃1βand IL⁃18 in lung tissue,and down⁃regulate the expressions of NLRP3,ASC,pro⁃caspase⁃1,cleaved caspase⁃1,GSDMD⁃N,p⁃c⁃Jun and c⁃Jun in lung tissue of mice.Conclusion Xuebijing can improve acute lung injury and inflammatory reaction of lung tissue induced by sepsis,and its mechanism may be related to down⁃regulating the expression of c⁃Jun and inhibiting its activation,thus inhibiting the activation of NLRP3 NLRP3 inflammasome and cell pyroptosis.
